The antibody reacts with active PAI-1, latent PAI-1 and t-PA-PAI-1 complexes. No cross reaction with PAI-2 or PAI-3. No effect on the functional activity of PAI-1. Can be used for immunodepletion of PAI-1 from human plasma.

Immunogen: PAI from human melanoma cell line

Monoclonal Antibody to Plasminogen Activator Inhibitor 1 (PAI-1)

Specificity: Binds to active PAI-1, latent PAI-1 and t-PA-PAI-1 complexes. No binding to PAI-2 or PAI-3. Interferes with the functional activity of PAI-1.

Derivation: Hybridization of NSO cells with spleen cells from a BALB/c mouse immunized with purified PAI-1 from the human melanoma cell line MJZJ according to the method of Wagner et al. (1).

Antibody class: IgG 2b

Purification: Purified from murine ascites by affinity chromatography on immobilized recombinant Protein A. The purity of the product was tested by Laemmli SDS-PAGE.

Application: For research use only:

- Immunoblotting

- Immunodepletion of PAI-1 from human plasma

- ELISA tests

Handling and  Storage: The antibody was lyophilized from a 1mg/ml solution in isotonic phosphate buffered saline, pH 7.4, containing 0.02% sodium azide and 20mg/ml mannitol. It is supplied in vials of 500µg and should be reconstituted with 0.5ml distilled water. For extensive dilutions a protein containing solution should be used (e.g. 1% bovine serum albumin in PBS). Lyophilized antibody should be stored at 2-8°C. Reconstituted antibody should be aliquoted and stored at -20°C or below. Avoid repeated freeze-thaw cycles.

Quantity: 500µg (lyoph.)

PAI-1 Actibind Test

-Solid phase EIA using a monoclonal anti t-PA antibody for immobilization of t-PA. The active site of t-PA remains accessible for binding of active PAI-1 present in the sample. For detection a peroxidase-conjugated monoclonal antibody specific for PAI-1 is applied.
