Since no one antibody works best for all applications (neutralization, blotting, ELISA, etc), we offer many different types of antibodies to help solve this problem. NFkB Antibodies

The transcription factor NFkB is widely recognized as a critical mediator of immune and inflammatory responses. In most cell types, NFkB is found in the cytoplasm where it is associated with an inhibitory protein known as IkB. An impressive variety of stimuli (tumor necrosis factor, interleukin 1, T-cell activation signals, bacterial endotoxins, viral transforming proteins, certain growth factors and reactive oxygen intermediates) lead to the rapid nuclear accumulation of NFkB by the induced phosphorylation and subsequent degradation of IkB. In the nucleus, NFkB regulates genes encoding cytokines, cytokine receptors, cell adhesion molecules, proteins involved in coagulation and genes involved in cell growth control. Additionally, NFkB is thought to be an important transcriptional regulator for HIV. Growing evidence indicates that the dysregulation of NFkB may be key to a number of diseases including arthritis and other inflammatory diseases, Alzheimer's disease, atherosclerosis and cancer.

NFkB is formed through the association of multiple subunits, either as a homodimer or heterodimer. Subunits have been identified as p50 (NFkB1), p65 (RelA), c-Rel, RelB and p52 (NFkB2). The classic NFkB form exists as a p50-p65 heterodimer and predominates in many cell types. Many of the possible combinatorial forms of homo- and heterodimers have been identified and growing evidence indicates that different forms of NFkB have different functions in cells. Interestingly, both the p50 and p52 subunits are derived from the precursor proteins p105 and p100 respectively, that each contain multiple copies of the so-called ankyrin repeat at their C-termini. Nuclear translocation of NFkB is confirmed by the use of electrophorectic mobility shift assays or by immunoblotting with nuclear extracts. The subunit composition of NFkB is confirmed by the use of antibodies that "supershift" the DNA/protein complex.

Specificity: antibody detects human p52 by immunoblotting only. Antibody may recognize p52 from other species. Recognition is at the N-terminal region of the p52/p100 subunit of NFkB.

IkB Antibodies

Three major forms of IkB-like molecules have been identified and each is characterized by multiple copies of ankyrin repeats. IkB-alpha and IkB-beta appear to be the major regulatory forms of IkB in most cells. These proteins interact with p65 or c-Rel containing forms of NFkB and block nuclear import by masking the nuclear localization sequences of NFkB. The activation of NFkB involves the inducible phosphorylation and subsequent degradation of IkB. Immunoblotting easily detects the hyper-phosphorylated forms of IkB-alpha, but not phosphorylated IkB-beta. Interestingly, IkB-alpha and IkB-beta mediate different NFkB responses. IkB-alpha appears to control more transient activation of NFkB in response to an inducer, while IkB-beta controls a persistent response. Bcl-3 interacts with p50 and p52 containing forms of NFkB, but rather than being an inhibitor it appears to function to stimulate transcription. The degradation of IkB is confirmed by immunoblotting.

Recognition is at the highly conserved C-terminal region. The antibody detects hypo- and hyper-phosphorylated forms of IkB-alpha. Antibody can be used for immunoprecipitation.  Western blot of IkB-alpha shows a 36 kDa band. RECOMMENDED DILUTIONS: 1:1000 dilution for western blot (band approx 36kda) recommend block with 5% dry milk for 2 hours. Perform all incubations except color development with TBS and 0.1% Tween-20 at room temp.
