DATA SHEET: mouse anti-Rat Nestin

Background: nestin is an intermediate filament protein that is abundantly expressed in neuroepithelial stem cells early in embryogenesis, but is absent from nearly all mature central nervous (CNS) cells. Nestin was originally defined as the antigen recognized by the monoclonal Rat401.

Antigen: paraformaldehyde fixed spinal cord from day 15 rat embroys.

Activity: -recognizes rat nestin (does not cross react with human nestin). (1)

              -western blot (2) doublet of approx 200kD, RAT- 401 positive cells are found throughout the

              developing neural tube, but not in the adult CNS. More than 90% of dissociated E11 neural tuvbe cells are rat-401 positive.

Mouse anti-Nestin Monoclonal ab (Human/Mouse/Rat)

Background: The cytoskeleton consists primarily of core structural proteins that include microfilaments, microtubules, and intermediate filaments (IFs). IFs contain more than 50 distinct proteins that are organized into six different subtypes: Type I/II keratin proteins expressed in epithelia cells, type III vimentin/desmin, type IV neurofilament proteins, type V nuclear lamins, and type VI nestin proteins expressed primarily in embryonic cells. Nestin has a conserved core region (amino acids 7 to 314), which contains an alpha helical domain that is involved in coiled-coil assembly of IFs. The C-terminal region of nestin is similar to type IV IFs, since it contains highly charged amino acids, many glutamate residues, and an 11 amino acid repeat motif. Nestin is expressed in the cerebrum during embryonic development, in the cerebellum during early postnatal development, and in dermatomal cells and myoblasts during myogenesis. In vitro, nestin forms homodimers and homotetramers, but not IFs, and can co-assemble with type III vimentin and type IV internexin proteins. Thus, nestin is a core IF protein that is essential for proper cytoskeletal formation during neurogenesis and myogenesis.

package: 50ug (200ul) or 150ug (600ul) purified antibody (0.25mg/ml) in a 50%glycerol, 20mM sodium phosphate pH7.5, 150mM NaCl, 1mg/ml BSA and 1.5mM sodium azide.

Use: Suitable for western blot on human, rat ,and mouse using approx 0.5ug/ml ab on total cell lysate. Major band at Mw 260kDa. Recommend 2-3 hour RT or overnight 4 DEG C incubation with enhanced chemiluminescence detection. Not suitable for immunofluorescence.

Storage: Store at -20 DEG C upon receipt. Avoid frequent freeze thaw cycles. Recommend aliquoting. Avoid frequent freeze thaw cycles.
