DATA SHEET:   Mouse anti-human MMP-9 monoclonal (histochemistry/WB)

Package Size: 100ug (lyophilized) with 100ug BSA. Recommend reconstituting the antibody with sterile PBS, pH 7.4 or sterile 20mM Tris saline (20mM Tris containing 0.15M NaCl), pH 7.4 to yield a 100ug/ml antibody concentration. Allow to reconstitute for at least 2 hours with gentle mixing prior to use.

Reactivity: Binds to human and guinea pig MMP-9 (rabbit and rat reported). Binds to epitope in amino acids 626-644 of human MMP9. Not reactive with mouse, bovine or hamster- reacts with both latent (92kda) and active (83kda) forms of MMP-9. Does not react with MMP-1, MMP-2, MMP-3 or MMP-13.

Suitable for: western blotting and histochemistry (frozen and paraffin sections)

Positive Control: conditioned medium from dexamethasone treated HT 1080 cells (stimulated with PMA). -incubate HT1080 cells for 2 hours at 37 DEG C in serum free media containing 1uM dexamethasone. Collect and centrifuge medium, concentrate 5- 20X . Store in aliquots at -20 DEG C.

Rabbit anti-MMP-9

Specificity: Binds to human MMP-9. No cross reactivity with the other MMP family members (MMP-1, MMP-2, and MMP-3). -also active on rat MMP-9 in cytochemistry on cultured rat C6 cells.

Immunogen: Synthetic peptide based on the human sequence.

Application: Western Blotting applications. When used against the reduced protein, it identifies both a 92 kDa and an 88kDa species (the pro-form and active form of the enzyme,respectively). The antibody does not bind as well to the non-reduced protein, and has limited use in immunoprecipitation,immuno-histochemistry, and ELISA applications.

MMP-9 is constitutively produced by some tumor cell lines (i.e., HT1080, HL60, and U937), but not in most quiescent cells and tissues. The phorbol ester TPA stimulates production of MMP-9 in some cell types. MMP-9 levels in culture media (pg/mL) are often below the threshold of detection by standard Western Blotting. The enzyme can be concentrated from culture media by gelatin-agarose affinity chromatography (Goldberg,G.l.et al.J.Biol.Chem. 267, 4583-4591. 1992). This can prevent appearance of a spurious band of antibody binding in the vicinity of 68 kDa displayed by some concentrated media.

Dilution: Western Blots is 1:1,000 when using colorimetric substrates such BCIP/NBT, and 1:5,000 for chemiluminescent substrates. Higher concentrations of antibody may be needed for non-human samples.

Optimal working dilutions must be determined by the end user.

Storage/Handling: The undiluted antibody solution is stable for approximately 6 months when stored between 2 and 8'C. For longer storage, mix antibody solution with glycerol 1:1 to prevent freezing, and store at -20'C. Diluted antibody can be used several times if not contaminated; store between 2 and 8'C with 0.01% sodium azide as preservative.

Background: It recognizes proteins of 92kDa and ~86kDa which are identified as pro (latent) and activated forms of matrix metalloproteinase 9 (MMP-9; also known as 92kDa collagenase IV or gelatinase B). Clone IIA5 Mab shows no cross-reaction with pro and active forms of other MMPs. MMPs are a group of enzymes involved in matrix degradation. They share some important characteristics, such as a common mode of activation, a conserved amino acid sequence in the putative metal binding-active site region, and inhibition by specific proteinase inhibitors known as tissue inhibitors of metalloproteinases (TIMPs). Degradation of the extracellular matrix (ECM) is an essential step in tumour invasion and metastasis. The MMPs each have different substrate specificities within the ECM and are important in its degradation. MMP activity is dependent on the levels of activated MMP and TIMPs.
