Since no one antibody works best for all applications (neutralization, blotting, ELISA, etc), we offer many different types of antibodies to help solve this problem. Matrix metalloproteinases (MMPs) are a family of enzymes that are responsible for the degradation of extracellular matrix components such as collagen, laminin and proteoglycans. In addition to sequence homology, all MMPs share the following characteristics: the catalytic mechanism is dependent upon a zinc ion at the active center, they cleave one or more extracellular matrix components, they are secreted as zymogens which are activated by removal of an approximately 10 kDa segment from the N-terminus and they are inhibited by tissue inhibitor of metalloproteinases (TIMP). These enzymes are involved in normal physiological processes such as embryogenesis and tissue remodeling and may play an important role in arthritis, periodontitis, and metastasis.

MMP-3 (stromelysin-1) is secreted as a 59/57 kDa zymogen which is proteolytically processes to the 45 and 28 kDa active forms. This enzyme degrades a wide spectrum of extracellular matrix proteins, including cartilage proteoglycans, fibronectin, type IV and IX collagen and laminin. MMP-3 is thought to play an important role in pathophysiological degradation processes associated with conditions such as rheumatoid arthritis and cancer cell invasion.

Localization: cytoplasmic

Activity: It recognizes one doublet of 54kDa/59kDa and another doublet of 44kDa/49kDa, which are identified as unglycosylated and glycosylated species of pro (latent) and active forms of matrix metalloproteinase-3 (MMP-3; also known as Stromelysin-1 or Transin). Clone SL-1 IID4 shows no cross-reaction with the pro and active forms of other MMPs. MMPs are frequently expressed in malignant neoplastic cells and play crucial roles in tumor invasion and metastasis. Tissue inhibitors of matrix metalloproteinases (TIMPs) control the activity of MMPS. The tissue distribution of three major MMPs has been defined in human mammary pathology: 1) collagenase (MMP-1) which degrades fibrillar interstitial collagens; 2) a 72-kDa gelatinase (MMP-2) which mainly degrades type IV collagen and denatured collagens; and 3) stromelysin (MMP-3) which has a wider range of action, degrading several matrix components including the core proteins of proteoglycans, laminin and non-helical regions of collagens.

Package Size: 100ug (lyophilized) with 100ug BSA. Reconstitute with 1ml sterile PBS pH 7.4 or sterile 20mM Tris-saline (20mM Tris containing 0.15 NaCl). Let set at least 2 hours at 4 DEG C. Mix well before using. If suitable for use, addition of 0.1% sodium azide is recommended.

Antigen: purified human pro-MMP-3 purified from the conditioned media of rheumatoid synovial fibroblasts.

Reactivity: Binds to human MMP-3 (Latent (59/57kda) and active (45/28kda)forms and can react with MMP3/TIMP complexes. The antibody has approx 34% cross reactivity with human MMP-10. (also pig, not with mouse, rat or  rabbit or bovine).

Suitable for: western blotting (1ug/ml)using chemiluminescence detection Recognizes both latent and active forms of MMP-3. ref#2

-paraffin sections (with trypsin pretreatment or high temp release) clone B in ref # 2 & 3 (for paraffin sections, staining can be enhanced by culturing tissue in 2uM monesin for 3 hours. Tissue may also be fixed with PLP (periodate-lysine-4% paraformaldehyde) and stained using 5-10ug/ml.

Positive Control: HT180 treated media (incubate HT1080 cells for 2 hours at 37 DEG C in serum free media containing 100nM PMA. Collect and centriguge medium, concentrate if necessary (store -20DEGC)

Storage: Store 4 DEG C or lower. Avoid frequent freeze thaw cycles. Antibody is stable for at least 6 months when stored frozen undiluted and up to 1 month at 4 DEG C when neat or diluted as per above recommendation. For prolonged storage add 0.02% thimerosal or 0.1% azide.

Rabbit Anti-MMP-3 (Stromelysin-1,Proteoglycanase)   (Carboxy terminal region)

Immunogen: Synthetic peptide based on the human sequence.

Specificity: The antibody binds to Stromelysin-1 and Stromelysin-2, but does not cross react with Stromelysin-3, nor to other MMP family members (MMP-1, MMP-2A, MMP-2B and MMP-9).

Applications: Anti-MMP-3 works well for Western Blotting applications and when used against the reduced protein identifies bands at 59 kDa and 57 kDa (the pro-form), as well as active forms. The 21 kDa active form is not recognized by the antibody. This antibody will also bind to the non-reduced protein, and thus has application in immunoprecipitation, immunohistochemistry, and ELISA. Stromelysin levels in quiescent cell cultures and tissues is minimal. Stimulation of synthesis or concentration of the protein from culture medium is often needed to visualize blotted bands. Cell types differ greatly in the quantity of Stromelysin produced. A recommended starting concentration for Western Blots in 1:1,000 when using colorimetric substrates such BCIP/NBT, and 1:5,000 for chemiluminescent substrates. Higher concentrations of antibody may be needed for non-human samples. Optimal working dilutions must be determined by the end user.

Storage: The undiluted antibody solution is table for approximately 6 months stored between 2 and 8'C. For longer storage, mix antibody solution with glycerol 1:1 to prevent freezing, and store at -20'C. Diluted antibody can be used several times if not contaminated; store between 2 and 8'C with 0.01% sodium azideas preservative.

Specificity: Binds to active and latent Stromelysin-1 and Stromelysin-2, but not Stromelysin-3 or other MMP family members (MMP-1, MMP-2, MMP-9).

Applications: Works well in Western Blotting identifying reduced protein bands at 59 Kda and 57 Kda (MMP-3 proenzyme), as well as all active forms of MMP-3. This antibody will also bind to the non-reduced protein, and can be applied in immunoprecipitation, immunohistochemistry, and ELISA. Stromelysin levels in quiescent cells and tissues is minimal. Some cells respond to mitogens (IL- 1A, TNF-a, TPA) by stimulating synthesis, but protein concentration from medium may be needed to visualize blotted bands.
