Package Size: 100ug (lyophilized) with 100ug BSA. Reconstitute with 1ml sterile PBS pH 7.4 or sterile 20mM Tris-saline (20mM Tris containing 0.15 NaCl). Let set at least 2 hours at 4 DEG C. Mix well before using. If suitable for use, addition of 0.1% sodium azide is recommended. Do Not add azide if to be used in live cell or neutralization experiments

Reactivity: Binds to human MMP-2. Binds to epitope in amino acids 524-539 of human MMP-2 (amino acids 468 to 483 of human MMP-2 using numbering from propeptide sequence). -also reacts with rat, mouse and bovine -reacts with both latent(72kda and active (66kda) MMP-2.)

Suitable for: western blotting (1ug/ml)and histochemistry (frozen and paraffin sections)

-recognizes both latent and active MMP-2. No pretreatment needed for paraffin sections.

Positive Control: conditioned medium from dexamethasone treated HT 1080 cells (stimulated with PMA).-incubate HT1080 cells for 2 hours at 37 DEG C in serum free media containing 1uM dexamethasone. Collect and centrifuge medium, concentrate if necessary. Store in aliquots at -20 DEG C.

Storage: Store 4 DEG C or lower. Avoid frequent freeze thaw cycles. Antibody is stable for at least 6 months when stored frozen undiluted and up to 1 month at 4 DEG C when neat or diluted as per above recommendation. For prolonged storage add 0.02% thimerosal or 0.1% azide.

Rabbit Anti-MMP-2   [Gellatinase-A](Hinge region)

Specificity: binds to human MMP-2 but does not cross react with MMP family members (MMP-1, MMP-2B, and MMP-3, MMP-9). -also recative in cytochemistry on rat C6 cells.

Immunogen: Synthetic peptide based on the hinge region of human MMP-2 sequence.

Applications: -Recognizes reduced and native MMP-2, free or bound to TIMP-2. Suitable for Western Blotting applications, immunoprecipitation, immunohistochemistry and Elisa. MMP-2 is constitutively produced in quiescent cells and tissues and the enzyme has a high specific activity against denatured collagen. The low protein content produced in cell culture (pg/ml levels) are often below WB detection. The enzyme can be concentrated by gelatin-agarose affinity chromatography (see J. biol Chem 267,4583-4591, 1992) or by immunoprecipitation.

Dilution: Starting dilution for Western Blots is 1:5,000 for chemiluminescent substrates. Higher concentrations of antibody may be needed for non-human samples. Strongly recommend use of enhanced chemiluminescence (ie, Amersham's ECL-Plus (tM) or Pierce Super-Signal ULTRA (tm) or equivalent).

Optimal working dilutions must be determined by the end user.

Storage: The undiluted antibody solution is stable for approximately 6 months when stored between 2 and 8'C. For longer storage, mix antibody solution with glycerol 1:1 to prevent freezing, and store at -20'C. Diluted antibody can be used several times if not contaminated; store between 2 and 8'C with 0.01% Sodium Azide as preservative.

Source: human recombinant MMP-2 purified from mammalian cells

Activity: A412/hour/ug protein within the range of 8.0-10.0 in standard thiopeptolide hydrolysis assays 14.

Prep: The proenzyme form of MMP-2 was activated by organo-mercurial compount, 4-aminophenyl mercuric acetate (APMA). APMA is removed through a G-25 column and purified active MMP-2 is formulated in buffer.

Use: This active MMP-2 enzyme may be used as a positive control or standard for zymographic analysis. Western blotting (approx 0.1ug/lane), or substrate cleavage assays. Titer for each use.

Storage: Store at -80'C. Product guaranteed until expiration date as stated. Avoid repeated freeze- thawcycles. Dispense and/or dilute original solution into working aliquots using siliconized vials and refreeze.

Purity: >95% by SDS-PAGE. A portion (less than 10%) of the enzyme may be seen as a naturally occurring dimer. Dimerization does not interfere with activation. During storage, a small portion (less than 10%) of the enzyme may also become activated.|
