RDI Divison of Fitzgerald Industries Intl  offers a wide line of  antibodies. Since no one antibody works best for all applications (neutralization, blotting, ELISA, etc), we offer many different types of antibodies to help solve this problem. Please inquire for other applications or types of antibodies not listed below.


Anti-MMP1 Antibodies (see below 2 monoclonals and 2 polyclonals)
DATA SHEET:  Mouse anti-human MMP-1 monoclonal (Active Form)   WB

Package Size: 100ug (lyophilized)with 100ug BSA.

Dilution: Tighten cap, tap vial to settle contents. Reconstitute with 1ml sterile PBS pH 7.4 or sterile 20mM Tris-saline (20mM Tris containing 0.15M NaCl) pH 7.4. Allow to set 2-3 hours at 4 DEG C, mix well. Addition of 0.1% sodium azide recommended or other preservative if suitable for your system (do not add azide if to be used in neutralization type reactions)

Antigen: amino acids 81-87 of human MMP1 ( FVLTEGN)

Reactivity: Binds to human MMP-1. Binds to active form of human MMP1 (not with the latent form).
Suitable for: western blotting (10ug/ml) band at   approx 42kDa..

Positive Control: conditioned medium from HT 1080 cells (stimulated with PMA for 2 hrs at 37 DEg C).-Collect and centrifuge medium, concentrate if necessary. Store in aliquots at -20 DEG C.

Storage: Store 4 DEG C or lower. Avoid frequent freeze thaw cycles. Antibody is stable for at least 6 months when stored frozen undiluted and up to 1 month at 4 DEG C when neat or diluted as per above recommendation. For prolonged storage add 0.02% thimerosal or 0.1% azide. Dilutions should be made in buffers containing at least 0.1% BSA .

-antibody be may reused multiple times when used in western blot (if suitable for your particular use), however, you should add preservative and run a positive control to assure reactivity).

Rabbit Anti-MMP-1

[Interstitial Collagenase; Fibroblast Collagenase](Hinge region)

Quantity: 50 ug of Peptide-affinity purified immunoglobulin. In 30 mM Tris pH 7.4, 0.5M NaC1, 0.05% Na azide.

Specificity: binds to MMP-1 but does not cross react with MMP family members (MMP-2A, MMP-2B, and MMP-3, MMP-9). This antibody is complementary to cat#RDI-MMP1Pabr recognizing the hemopexin domain of MMP-1.

Immunogen: Synthetic peptide based on the hinge region of human MMP-1 sequence.

Applications: -western Blotting appl;ications. When used against the reduced protein, it identifies bands at 53 kDa and 51 kDa (pro-enzyme), as well as active forms. The antibody also binds the non-reduced protein, and therefore can be used in immunoprecipitation, immunohistochemistry, and Elisa.

Note: Collagenase levels in quiescent cells and tissues is minimal. Stimulation of synthesis or concentration of enzyme from culture medium is often needed to visualize bands on Western Blots. Cell types differ greatly in the quantity of Collagenase produced.

Dilution: Starting dilution for Western Blots is 1:1,000 when using colorimetric substrates such BCIP/NBT, and 1:5,000 for chemiluminescent substrates. Higher concentrations of antibody may be needed for non-human samples.
Optimal working dilutions must be determined by the end user.

Storage: The undiluted antibody solution is stable for approximately 6 months when stored between 2 and 8'C. For longer storage, mix antibody solution with glycerol 1:1 to prevent freezing, and store at -20'C. Diluted antibody can be used several times if not contaminated; store between 2 and 8'C  with 0.01% Sodium Azide as preservative.

Rabbit Anti-MMP-1

[Interstitial Collagenase; Fibroblast Collagenase]    (Hemopexin domain)

Quantity: 100 ug peptide-affinity purified immunoglobulin. with sodium azide or thimerosal as preservative.

Specificity: Binds to MMP-1 but does not cross react with the other MMP family members (MMP-2, MMP-3, MMP-9). This antibody is complementary to cat#RDI-MMP1Habr which recognizes the hinge region of MMP-1.

Immunogen: Synthetic peptide based on the hemopexin domain of human MMP-1 sequence.

Applications: Western Blotting applications. When used against the reduced protein, it identifies bands at 53 kDa and 51 kDa (proenzyme), as well as initial active forms. The hemopexin domain is cleaved from the MMP-1 zymogen to produce its ultimate active forms, which are not recognized by this antibody (cat#RDI-MMP1Pabr). The antibody also binds the non-reduced blotted protein, and may be useful for immunoprecipitation, immunohistochemistry, and ELISA.


Note: Collagenase levels in quiescent cells and tissues is minimal. Synthesis can be stimulated in receptive cells by TPA or TNF-a, but concentration of enzyme from culture medium may be needed to visualize bands on Western Blots. Rodent species appear to lack MMP-1.

Dilution: Starting dilution for Western Blots is 1:1,000 when using colorimetric substrates such BCIP/NBT, and 1:5,000 for chemiluminescent substrates. Higher concentrations of antibody may be needed for non-human samples. The immunogen peptide sequence is well- conserved among species. Optimal working dilutions must be determined by the end user.

Storage: The undiluted antibody solution is stable for approximately 6 months when stored between 2 and 8'C.
