Matrix metalloproteinases (MMPs) are a family of enzymes that are responsible for the degradation of extracellular matrix components such as collagen, laminin and proteoglycans. In addition to sequence homology, all MMPs share the following characteristics: the catalytic mechanism is dependent upon a zinc ion at the active center, they cleave one or more extracellular matrix components, they are secreted as zymogens which are activated by removal of an approximately 10 kDa segment from the N-terminus and they are inhibited by tissue inhibitor of metalloproteinases (TIMP). These enzymes are involved in normal physiological processes such as embryogenesis and tissue remodeling and may play an important role in arthritis, periodontitis, and metastasis.

MMP-13 (Collagenase-3) is secreted as a 60 kDa zymogen which is proteolytically processed to the 48 kDa active form. MMP-13 shows substrate specificity toward interstitial collagens I-III collagen and gelatin, preferentially cleaving type II collagen over type I and III. MMP-13 may be especially important in the turnover of articular cartilage, which is rich in type II collagen. Active MMP-13 can be inhibited by TIMP-1, TIMP-2, and TIMP-3. Numerous studies have shown a correlation between collagenase expression and metastatic potential and suggest that it may be useful marker for the diagnosis or prognosis of cancer.

Positive Control: expressed NSO cells

Storage: Store 4 DEG C or lower. Avoid frequent freeze thaw cycles. Antibody is stable for at least 6 months when stored frozen undiluted and up to 1 month at 4 DEG C when neat or diluted as per above recommendation. Recommend adding 0.1% azide if sutiable for your assay system.

Human collagenase-3 (MMP13) is a recently identified member of the matrix metalloproteinase (MMP) family that is expressed in breast carcinomas and in articular cartilage from arthritic patients. The MMP13 gene has been isolated and characterized. This gene is composed of 10 exons and 9 introns and spans over 12.5 kb. The overall organization of the collagenase-3 gene is similar to that of other MMP genes clustered at chromosome 11q22, including fibroblast collagenase (MMP-1), matrilysin (MMP-7), and macrophage metalloelastase (MMP-12), but is more distantly related to genes coding for stromelysin-3 (MMP-11), gelatinase-A (MMP-2), and gelatinase-B (MMP-9), which map outside of this gene cluster.
