Reactivity: The monoclonal antibody reacts with virtually all cells of the
myeloid lineage (from myekloblast to neutrophils and monocytes). The monoclonal
antibody does not react with human lymphocytes, thrombocytes and erythrocytes.
96% of all AML's are found positive; 100% of all ALL's are found negative.
Applications: Determination of Acute Myeloid Leukaemia.
Methods: Indirect immunofluorescence staining with analysis by FACS or
fluorescence microscopy.

Source: Ascites fluid of tumor bearing BALB/c mice.
Purification: Ammoniumsulphate precipitation and ion exchange chromatography.
Packing: Each vial contains 1 ml with approximately 0.1mg/ml monoclonal
antibodies and 10 mg BSA in 20 mM TRIS, 150 mM NaCl, pH 8.0.
Preservative: Merthiolate (0.001%).
Storage: Monoclonal antibodies should be stored below -18Deg C. Repeated
thawing and freezing may reduce the antibody activity and should be avoided. Do
not use this product beyond the stated expiration date.

Purification: delipidation and defibrination.
USE: ELISA (approx 1:120K)
western blot-approx 1:5000 for WB. detects multiple bands corresponding to 53kda and 15kda polypeptides and chain combination(68kda and 106kda).
Specificity: Reacts with myeloperoxidase from white cells. Cross reactivity will occur against myeloperoxidases from other species and tissues.

-suitable for use as coating antigen in an anti-MPO autoantibody immunoassay
-Anti-neutrophil cytoplasm antibodies (ANCA) are serological markers for
certain systemic vasculitides. These autoAbs have been subclassified into
cytoplasmic (cANCA) and perinuclear (pANCA) specificities according to the
indirect immunofluorescent staining pattern of ethanol-foxed neutrophils.
The major pANCA target was subsequently shown to comprise a myeloperoxidase
(MPO), an enzyme contained inalpha or primary azurophilic granules. The MPO
protein (MW ca 140Kda is a covalently linked complex of two alpha chains (MW ca
60kDa) and two beta chain (MW ca 14kDa). The purified MPO antigen can be used
for the detection of anti-MPO autoantibodies in immunoassays. The anti-MPO
autoantibodies are associated with the wide range of systemic vasculitides, eg.
periarteritis nodasa, microscopic polyarteritis,
pulmonary eosinophilic granulomatosis (Churg-Strauss syndrome). Low to moderate
anti-MPO autoantibodies are also reported in rheumatoid arthritis.
