Background: 5-Lipoxygenase (5-LO) is the initial enzyme that converts arachidonic acid to leukotrienes, important metabolites during inflammation and vasoconstriction. 5-LO activation occurs in response to a number of stimuli, such as differentiation and allergen challenges. The 5-LO gene-abundantly expressed in placenta, lung, and leukocytes-encodes for a protein of 674 amino acids with an apparent MW of 78kDa. 5-LO is a CA++ and ATP-dependent enzyme that, once activated, translocates from the cytosol to either a nuclear or plasma membrane compartment. A proline-rich domain between amino acids 566 to 577 of 5-LO has recently been identified as a binding site for the PTyr-binding protein Grb2, therefore linking tyrosine kinases with activation and redistribution of 5-LO. Further, tyrosine kinase inhibitors increased the activity of 5-LO and blocked the enzyme's subcellular redistribution.

antigen: a protein fragment corresponding to amino acids 442-590 from human 5-lipoxygenase.

Use: Suitable for western blot on human, rat, chick and mouse under using approx 1ug/ml a on total cell lysate of chick embryo fibroblast lysate. Major band =Mw 79 kDa

Storage: Store at -20 DEG C upon receipt. . Recommend aliquoting. Avoid frequent warming cycles.

Chick Embryo Firboblast (SL-29) Lysate: Derived from a chicken embryo fibrobblast cell line. The cell lysate is provided at a concentration of 1mg/ml in SDS-Page buffer (62nm Tris pH 6.8, 2% SDS, 0.9% B-mercaptoethanol, 0.003% bromophenol blue and 5% glycerol). Use approx 5-10ul for western blotting (when using enhanced chemiluminescence detection). Store -20 DEG C. DO NOT BOIL OR DILUTE.
