Specificity: Hemoxygenase (HO) is a microsomal enzyme that catalyzes the
oxidation of heme to the antioxidant molecules, biliverdin and carbon monoxide.
HO consists of two homologous isozymes, an inducible HO-1 and a constitutively
expressed HO-2. This rabbit antibody is specific for the HO-1 in human, rat,
monkey and bovine samples and does not cross react with HO-2.
In western blots, a single band at 32kDa is seen after stress induction.
In immunohistochemical studies, partial trypsin digestion is required of
the section prior to localization. This antibody can be used in formalin
fixed paraffin embedded sections.

Optimal dilutions must be obtained for all applications.
Storage: -20 C. Avoid repeated freeze/thaw cycles.

Background: Heme Oxygenase 1 and 2 (HO-1, HO-2) cleave the heme molecule
resulting in the production of carbon monoxide and bilverdin. HO-1 is a 288
amino acid monooxygenase with a molecular weight of 32kDa. While HO-2 is
constitutively expressed in tissues, HO-1 is rapidly induced by several stimuli
such as glutathione depletion, hemin, heat shock, heavy metals, oxidative
stress, oxidized LDL, anoxia and endotoxic shock. Similar conditions that
increase NO also induce HO-1 expression and activity. This suggests that
NO synthases, COX-2, and HO-1 are part of a general mechanism against stress
and pathogen invasion.

Storage: Store at -20 DEG C upon receipt. . Recommend aliquoting.
Avoid frequent warming cycles.

Background: Heme Oxygenase (HO) mediates the degradation of heme into biliverdin,
free iron, and carbon monoxide (CO). HO exists as two isozymes, designated
HO-1 and HO-2. These two isoforms share about 40% amino acid similarity including
several stretches of highly conserved sequences with matched predicted secondary
structure. HO-1, the inducible form, is found primarily in spleen and liver
Kupffer cells. However, HO-2 is constitutively expressed in abundance in
brain, testis, and liver parenchymal cells. Although HO-1 has an established
physiological role in heme catabolism, the HO-2 mechanism is unclear. However,
the HO-2 heme binding pocket is similar to that of HO-1 and heme degradation
is thought to proceed by similar mechanisms. In the brain, HO-2 colocalizes
with neuronal nitric oxide synthase (nNOS) within adventitial neurons and
in autonomic ganglia. Like the neurotransmitter NO, CO produced by HO-2
stimulates guanylyl cyclase. Also, HO-2 may function to limit acute and chronic
inflammatory responses. Thus, HO-2 serves as a mediator of heme degradation
and, in doing so, may generate molecules important for neurotransmission
and possibly immunomodulation.
