Description: Fibronectin is a dimeric glycoprotein which is present in cells, extracellular matrix, and blood.1 It possesses at least four binding sites for collagen, glycosaminoglycans, transglutaminase, and a cell surface receptor. Fibronectin is involved in cell adhesion, tissue organization, and wound healing. Comments: Mab   reportedly exhibited a characteristic fibrillar   pattern of cell surface labeling. Clone HFN 36.3 Mab reacts with both the plasma and cellular forms of fibronectin.

The National Institute of Occupational Safety and Health has issued a bulletin citing the potential explosion hazard due to the reaction of sodium azide with copper, lead, brass, or solder in the plumbing systems. Sodium azide forms hydrazoic acid in acidic conditions and should be discarded in a large volume of running water to avoid deposits forming in metal drainage pipes.

Comments: Mab cloneTV-1 reacts with the cellular as well as plasma form of fibronectin. Reportedly, after iv administration, Mablocalizes to tumor vessels where it binds to the underlying basement.1 Epitope recognized by this clone is not accessible in normal tissues to the circulating MAb indicating that it can be used to specifically target tumor vessels in vivo.1 It is reportedly useful for delivering vasoactive agents to tumors to induce increased vascular permeability or blood flow prior to treatment with chemotherapeutic drugs or MAbs

Activity: This Mab is directed against the peptide core and reacts with both the plasma and cellular forms of fibronectin.1 It blocks the fibronectin-medicated cell attachment not by disrupting the collagen-fibronectin interaction, but by interfering with the attachment of fibronectin to its receptor on the cell surface 1 .

Background: Fibronectin, a 240 kDa dimeric extracellular matrix protein, is secreted by mesenchymal cells and assembled into insoluble matrices. Fibronectin may precipitate in early steps of intracellular signal transduction by triggering responses to cell interactions with the extracellular matrix. Fibronectin exhibits different isoforms derived from alternative splicing, and variation in glycosylation. Cell adhesion, spreading , and focal formation are promoted through ligand binding of fibronectin to the fibronectin receptor, A5B1 integrin. TGFB has been shown to modulate expression offibronectin.

Immunogen:Fibronectin was purified from human plasma by binding to a denatured gelatin column followed by elution with high concentrations of arginine. The eluted matrial was further purified by gel filtration. Immunization occurred after single-band purity was assessed by SDS-PAGE.
