Background: FAK (focal adhesion kinase) is a novel protein tyrosine kinase
that co-localizes with integrins in cellular focal adhesions. It is a major
phosphotyrosine-containing protein in normal avian and rodent fibroblasts,
but exhibits enhanced tyrosine phosphorylation in cells transformed by Rous
sarcoma virus. Sequence analysis of cDNA encoding p125 FAK has revealed it
to be the prototype of a new class of protein tyrosine kinases that is part
of an integrin-regulated signal transduction pathway. FAK does not contain
SH2 or SH3 domains, but it does possess a 159 amino acid carboxy terminal
sequence (Focal Adhesion Targeting sequence) responsible for directing its
cellular localization. p125 FAK is phosphorylated in response to fibronectin,
as well as to extracellular adhesion matrix ligands. It appears that FAK
may be primarily autophosphorylated and this modification may affect the
interaction of FAK with other proteins. It has recently been shown that FAK
can associate with pp60(src) in RSV transformed cells and with c-fyn in normal
chicken embryo cells.

Use: Suitable for western blot on human, dog, rat, chick and mouse under
using approx 0.5ug/ml a on total cell lysate of human endothelial lysate.
Major band =Mw 125kDa. Also suitable for histochemistry and immunofluorescence
and immunoprecipitation (denaturing conditions)
Storage: Store at -20 DEG C upon receipt. Recommend aliquoting. Avoid frequent warming cycles.
