Rabbit anti-Rat Dopamine Transporter (DAT)   (IgG fraction) RT/MS Hu/Dog

Presentation: 100ug affinity pure antibody (lyophilized from 1mg/ml 100mM Tris pH7.5, 0.2% BSA and 0.05% sodium azide).

Reconstitution: Add 100ul sterile DI water, let set at least 30 minutes at 4 DEG C. Gently mix well. If desired, centrifuge 1-2 minutes at 1000 rpm to concentrate material in vial. Additional dilutions should ideally be made in buffers containing 0.1-0.2% BSA or other suitable carrier.

Purification: peptide affinity purified.

Antigen: An 18 amino acid peptide sequence near the amino terminus of rat brain dopamine transporter (assumed to be a cytoplasmic domain) with a Cysteine group added at the amino terminus and conjugated to KLH

Reconstitution: All peptide/antibody received in solution or reconstituted from Lyophilized vials should be stored frozen at -20oC or below in suitable aliquots. It is not recommended to store diluted solutions. Avoid repeated freeze and thaw.

Specificity: Predicted cross reactivity based on antigen sequence homology as indicated below:

Antigen sequence homology: 18 amino acid

-rat/mouse (100%; 18/18)

-bovine/monkey (77%, 14/18)

-human (72%) 13/18)

USE: Western Blotting (1-10 ug/ml for affinity pure using Chemiluminescence technique).

-band approx 80Kda against rat striatum but not cerebellum. Rat DAT contains various carbohydrate moieties and neuraminidase and N-Glycanase treatment reduces its mol weight to approx 55kDa. brain

ELISA (1:10K+; using 50-100ng UCP1/well.

Histochemistry & Immunofluorescence:Not tested:We recommend the testing of affinity purified antibody at 2-20ug/ml on formaldehyde fixed tissue.

Generally titer antibody to maximum usable dilution and then pretreat antibody with 10X weight of peptide for 2-3 hours RT or overnight 4 DEG C, then proceed with staining.
