Reactivity: human, monkey, mouse, rat and dog (others not tested)


Uses:

-Western blot: 1ug/ml for 2 hours RT

-histochemistry (methacarn and formalin/paraffin sections) approx 1ug/ml for 60 minutes RT

-for paraffin sections, requires boiling tissue sections in 10mM citrate buffer pH6.0 for 10-20 minutes followed by cooling at RT for 20 minutes.

-immunoprecipitation: use Protein A , Mab 2ug/mg (co-precipitates Cdk4)

-flow cytometry

-immunofluorescence

-Biological blockage (use azide free material)

Positive Control: Mantle cell lymphomas and breast carcinomas

Background: Cyclin D2 is a G1 cyclin required for G1-phase progression and is a strong candidate for a proto-oncogene. cyclin D2 can phosphorylate pRB when associated with cdk4 and/or cdk6. Mab from clone DCS-3.1 is particularly well suited for kinase assays

Specificity                                 DCS-6 reacts specifically with cyclin D1 protein present predominantly in G1 phase of cell cycle; it does not cross-react with D2 or D3 cyclins (the closest relatives within the cyclin family); mab DCS-6 is especially useful in tumor diagnosis: it can be used as fast method to identify cases with 11q13 amplifications. Cyclin D1 gene is identical to the BCL-1 and the PRAD-1 oncogene and is part of the chromosome 11q13 amplicon which is amplified in several common tumor types like breast, lung, esophagus, urinary bladder and head and neck carcinoma.
