PRESENTATION
:The monoclonal is presented at a concentration of
200µg/2ml in phosphate buffered saline containing 10mM sodium azide and 1mg/ml bovine serum albumin. We recommend that
each laboratory determine an optimum working titre
for use in its particular application.
STORAGE
For use within 1 month of purchase store at +4oC, for long term storage
aliquot antibody into small volumes and store at -20oC.

SPECIFICITY:
The antibody recognizes fibronectin in
frozen sections of human tissues. Specifically it stains the extracellular
adhesive glycoprotein in connective tissues and blood vessels. CBL 248 does not
recognize the soluble dimeric form of fibronectin (plasma fibronectin)
but stains strongly matrix fibronectin in tissues.
Animal biodistribution experiments have shown that
after intravenous administration, this antibody localizes to tumor vessels
where it binds to the underlying basement membrane. The antigen recognized is
not accessible in normal tissues to circulating antibody indicating that this
antibody can be used to target tumour vessels
specifically in vivo.

Description/Specificity
Mab 568 reacts with the cell binding part of the fibronectin molecule. Fibronectin
seems to play a key role in the tumor cell extracellular matrix during invasive
tumor growth. Increasing expression of stromal fibronectin has been reported during tumor progression. In immunohistochemistry the antibody is reactive with an
extracellular matrix glycoprotein in vessels and connective tissue.

Frozen sections
Species: recognizes fibronectin in human, monkey, rat
and hamster
Specificity: On frozen sections, this antibody only stains stromal
or connective tissue regions abundant in fibroblasts. In cultured cells, this
antibody only reacts with fibronectin.
Storage: Store at 4 DEG C (short term,) -20 DEG C long term. Avoid frequent
freeze thaw cycles. Recommend adding 0.1% sodium azide
or other preservative.
