-immunogen: partially purified rat camKinase II

-reactivity: rat, mouse, bovine, chicken and frog

-use: western blot (positive control rat or mouse brain tissue) and cytochemistry

-the antibody detects a 50kDa band corresponding to the expected mw of camKinaseII alpha isoform on immunoblots. Both the phosphorylated and non-phosphorylated forms are detected. Expected reactivity with the alpha, beta and gamma isoforms but not the beta isoform.

-the antibody detects a 60kDa band corresponding to the expected mw of camKinaseII on immunoblots. Since the sequence of the rat alpha, delta and gamma isofroms are conserved over this amino acid region, the antibody is expected to recognize the alpha, delta and gamma forms (not expected to reract with the beta isoform).

Immunogen: 14 amino acids synthetic peptide based on rat multifunctional calmodulin-dependent kinase 2 delta isoform (residues 7-20 coupled to KLH) -CTRFTDEYQLFEEL

Reactivity: rat/mouse (human and bovine with lesser reactivity)

Activity: detects an approx 50-60kDa band in western blots of crude rat brain and liver tissue extracts which corresponds to rat camkinaseII alpha subunit.

-As the sequences of the rat alpha,delta and gamma isoforms are conserved over this amino terminus region, this antibody is expected to recognize these isoforms. It is not expected to cross-react with the Beta isoform. In tissue extracts, this antibody recognizes a number of other bands in addition to these various isoforms.

Storage: 1 year at -20'C. At first thaw, tighten cap, centrifuge 1 minute 1000 rpm to concentrate antibody in vial. Recommend aliquoting, avoid frequent freeze thaw cycles.

Background: Calmodulin-dependent protein kinase II, a 50-60kDa serine/thronine kinase is also known as the multifunctional calcium/calmodulin dependent protein kinase, type II Cam-dependent PK or Cam-dependent multiprotein kinase. This kinase is present in many different tissues, but is found most abundantly on synapticvesicles in braincells. Different subunits of Cam kinase II have been described with apparent molecular masses of 54-60kDa. Alpha, B, gamma and delta are products of different genes, while Beta is an alternatively spliced product of the Beta gene. CaMKII exists as a homodimer of 6-12 subunits whose ratio varies depending upon the tissue localization of the kinase. Each subunit has an ATP-binding domain (arginine- A-A-serine/threonine) consensus phosphorylation site and a catalytic domain. A centrally located regulatory domain has calmodulin binding activity.

Reactivity: rat/mouse/bovine/frog/chicken/broadly with mammalian species for alpha isoform

Activity: detects an approx 50kDa band in western blots of crude rat, bovine and mouse brain tissue extracts which corresponds camkinaseII alpha subunit. -The antibody reacts with the alpha isoform in all mammalian cells plus chick and frog. Both the phosphorylated ad non- phosphorylated forms are recognized. Expected reactivity with the alpha, delta and gamma isoforms but not the beta isoform.
