Mouse anti-Calpastatin

Immunogen: Bovine skeletal muscle calpastatin.

Epitope: This antibody recognizes an epitope found between amino acid residues 295-501 (domain II) of human calpastatin.

Applications : Immunocytochemistry 1:1,000  

                      Western Blot 1:5,000

-detects calpastatin from bovine, human, pig and rat tissues and cells. This antibody does not cross-react with calpains or calmodulin. -has been successfully used in Western blot and immunocytochemical procedures. By Western blot, this antibody detects 150, 125, 90 and 70 kDa proteins which have been suggested to be proteins translated from different start sites of the calpastatin gene. Immunofluorescence staining of calpastatin in porcine LLC-PK1 cells results in diffuse cytoplasmic staining and weak nuclear staining.

Background: The calpain system has been detected in every vertebrate tissue examined, and has been suggested to play a regulatory role in cellular protein metabolism. This regulatory role may have important implications in platelet aggregation and pathologies associated with altered calcium homeostasis and protein metabolism such as ischemic cell injury and degenerative diseases. Inhibitorscontinued of calpain have been shown to block dexamethasone and low-level irradiation induced apoptosis in thymocytes suggesting that calpain has a regulatory or mechanistic role in apoptotic cell death.

Calpastatin, a specific endogenous inhibitor of calpain, has a predicted molecular weight of ~77 kDa and an internal repeat of four homologous domains which allow it to inhibit multiple calpain molecules simultaneously. It functions by binding to calpain when calpain binds calcium. When calpastatin is subjected to proteolytic digestion, fragments of calpastatin as small as 15 kDa still retain inhibitory activity.

Description: The protein consists of a single 70Ka protein. Calpastatin was purified by heat precipitation of other proteins followed by AFF-GEL Blue chromatography.This protein can be used for absorption tests and immunoblots.

Storage: Freeze in small aliquots (approx 1ug) and keep at -20 DEG C. Avoid repeated freezing and thawing.

Use: -Dilute 1ul of anti-calpastatin antiserum in 1ml of the usual buffer for immunohistochemistry

(final dilution 1:1000).

-Add 1ug of protein to the 1ml of diluted antibody solution and mix well.

-Incubate for at least 6 hours in the cold

-Apply to tissue sections and incubate for 3 days

-Complete the immunohistochemical reaction as usual (biotinylated second antibody, ABC-complex DAB etc).

-As a result, the immunostaining should be strongly reduced or even completely prevented.

Description: Calpastatin is an endogenous protease inhibitor that acts specifically on Calpain I, a CA 2+ dependent cysteine protease. Widely distributed in mammalian and avian cells. This recombinant calpastatin corresponds to domain I of human calpastatin. The inhibitory sequence of human domain I consists of at least 18 amino acids residues.

Purity: homogeneous by SDS-PAGE

MW: 14kDa

Solubility: DI water and aqueous bufers

Storage: After reconstitution, Freeze in working aliquots and keep at -20 DEG C. Avoid repeated freezing and thawing.
