RDI Division of Fitzgerald Industries Intl  offers a wide line of  antibodies. Since no one antibody works best for all applications (neutralization, blotting, histochemistry, ELISA, etc), we offer many different types of antibodies to help solve this problem. Monoclonal anti-human MHC Class I (HLA-A, B and C)

cat# RDI-MHC1abm-3, new cat#: 10R-M109a   $346.50/100ug

Presentation: 100ug in 0.1ml (1mg/ml)50 mM Sodium Phosphate pH 7.5, 100 mM Potassium Chloride, 150mM NaCl, and 0.5mg/ml Gentamicin Sulfate.

Clone: 3F10

Isotype: Mouse IgG2a

Immunogen: Human HSB-2 T cell line

Reactivity: Antibody 3F10 recognizes a monomorphic Class I epitope on the a chain. Antibody 3F10 (whole molecule) is cytotoxic in the presence of complement (1).

STORAGE: Store at 2 - 5oC. Freeze/Thawing not recommended.

Derivation: Antibody from (low FBS containing) tissue culture supernatant was Protein A purified to >95% mouse immunoglobulin by SDS-PAGE (<1% bovine immunoglobulin).

PERFORMANCE: 500K cultured HPB-MLT cells were washed and incubated 45 minutes on ice with 80 ul of anti-MHC Class I at 10 ug/ml. Cells were washed twice and incubated with 2o reagent Goat anti-mouse IgG:FITC, after which they were washed three times, fixed and analyzed using a BD FACS Calibur. Cells stained positive with a mean shift of 2.26 log10 fluorescent units when compared to a Mouse IgG2a negative control (RDI- MIGG2AX2 $188.00) at a similar concentration.

Background: Human MHC Class I (HLA-A, B & C) molecules are expressed on nearly all nucleated cells. MHC Class I molecules consist of heterodimers of a highly polymorphic transmembrane a chain and an invariant b chain. MHC Class I molecules bind to endogenously synthesized peptides of about 8-10 amino acids which are presented to CD8+ T cells. Recent evidence now indicates exogenously derived peptides are also presented by MHC Class I molecules.

PERFORMANCE: 500K cultured HPB-MLT cells were washed and incubated 45 minutes on ice with 80 ul of anti-MHC Class I/FITC at 1:50 dilution. Cells were washed three times, fixed and analyzed using a BD FACS Calibur. Cells stained positive with a mean shift of 1.66 log10 fluorescent units when compared to a Mouse IgG2a/FITC negative control (RDI- MIGG2AFTX2 $188.00) at a similar concentration. Binding was 95% blocked when pre incubated 10 minutes with 20 ul of 0.5 ug/ml anti-MHC Class I antibody (unconjugated, cat#RDI-MHC1abm-3f $312.00/100ug)

Cells were washed twice and incubated with 2o reagent Streptavidin/ R-Phycoerythrin), after which they were washed three times, fixed and analyzed using a BD FACS Calibur. Cells stained positive with a mean shift of 2.92 log10 fluorescent units when compared to a Mouse IgG2a/Biotin negative control (RDI- MIGG2aBTX $188.00) at a similar concentration. Binding was 98% blocked when pre incubated 10 minutes with 20 ul of 0.5 ug/ml anti-MHC Class I antibody (unconjugated, cat#RDI-MHC1abm-3 $312.00/100ug)
