RDI Division of Fitzgerald Industries Intl offers a wide line of antibodies. Since no one antibody works best for all applications (neutralization, blotting, histochemistry, ELISA, etc), we offer many different types of antibodies to help solve this problem. Purification: Protein A sepharose
Clones(isotype) 3A1, 1G4,
1D6 or 1D3 (all mIgG1 kappa)
derived from hybridization of X63-Ag8-653 myeloma
cells with spleen cells of a Balb/c mice immunized
with purified human proinsulin.
Specificity: -Both clones are directed to different antigenic determinants.
-Clone 3A1 reacts with the site of connection of C-peptide and C-end of B-chain of proinsulin molecule.
Clones 1D6 and 1D3 Mab react with different epitopes on human proinsulin molecule, both of each are generated by the interactions between insulin and C-peptide parts of pro-hormone.
-Kd for clone 3A1 is 1.1 X 10 E-08M
-Kd for clone 1G4 is 2.6 X 10 E-07M
-Kd for clone 1D6 and 1G4 >3 X 10 E-08M
-These clones do not cross react with human, bovine, porcine insulin or bovine and porcine proinsulin or human c-peptide.
Application: Detection of human proinsulin in two site enzyme immunoassay.
Assay recommendations:

(Cat#RDI-TRK2IP10-D6C4, new cat#: 10R-I136a or RDI-TRK2IP10-D3E7, new cat#: 10R-I136b - $412.50/mg) for labeling.
- 1D6 or 1D3 Mabs are used for direct capture prohormone from serum sample and insulin-specific 7F8, D6C4 or D3E7 Mabs (Cat# RDI-TRK2I1-7F8, new cat#: 10R-I134c, RDI-TRK2IP10-D6C4 or -D3E7) as a conjugates.
- Construction of a sensitive two-site sandwich enzyme-linked/RIA immunosorbent assays for the measurement of intact human proinsulin in serum or plasma with the detection limit in buffer 0.4 - 0.5 pmol/L.
- Specific immunosorbtional (immobilized 1D6/1D3 Mabs) removal of proinsulin molecules from serum samples before the measurement of insulin C-peptide.
Clones 3A1 and 1G4 can be used in immunohistochemistry on frozen sections.
Safety: Although this antibody contains low amounts of sodium azide (0.1%), appropriate care must be taken when using this product. Avoid use of metal drains and flush with copious amounts of water.
Note:Upon receipt, tighten cap and centrifuge for 1-2 minutes at 1000 rpm to concentrate antibody in bottom of vial. If desired, can add PBS with 0.1% sodium azide to bring concentration to 1mg/ml.
