Description: Non-insulin-dependent diabetes mellitus (NIDDM) is a multifactoral disease with both environmental and genetics causes. Genome-wide screening procedures have identified several susceptibility loci for NIDDM within the human genome. A putative sugar transporter that has been localized to human chromosome 20q12-q13.1, one of the genomic loci associated with NIDDM. Because of the strong resemblance of this novel protein to members of the mammalian facilitative glucose transporter family (GLUT), the protein is known as GLUT10 (HGMW-approved gene symbol SLC2A10). Data suggests that GLUT10 an excellent candidate for a susceptibility gene involved in NIDDM.
Host Species: Rabbit
Specificity: Residues with residues 367-385 (ILSTAKKTKPHPRSGDPSA) of the human GLUT10 protein.
Applications: Western blot - Use at 1:500 to 1:1,000 dilution. However, the investigator should determine the optimal dilution for a specific application such as immunohistochemistry.
Format: Affinity purified IgG
Storage: Maintain at +2-8°C for 3 months or at -20°C for longer periods. Stable for 1 year. Avoid repeated freeze-thaw cycles.

Activity: Reacts only with human Glut 3, does not cross react with Glut 3
from rat or mouse. Exhibits only a single band of Mr 47Kda on western blot.
Useful for study of Glut 3 in human brain and specific human tumor cell lines.
Use: -Western blot: approx 1:5K dilution against whole human brain homogenates.
-Dot blot: 1:5K against peptide-BSA.
-immunocytochemistry:1:500 against human intestinal tumor cells.
Antigen: A 12 amino acid synthetic peptide corresponding to the carboxy terminus
of the human GLUT 3 sequence ( SIEPAKETTNV) conjugated to keyhole limpet
hemocyanin via a cysteine on the amino terminus of the peptide.

Activity: Stains Glut 5 as found in homogenates from Caco-2 human
intestinal cell lines. Does not cross react with Glut 1, 2, 3 or 4 from human,
mouse or rat.

kidney; no reaction against brain, gut or muscle from rat. Does not react
with human blood, fat, liver or muscle. Also reacts with testis, human intestinal

Glut isoform differ in their tissue expression, substrate specificity and kinetic characteristics. Glut-1 mediates glucose transport into red cells, and throughout the blood brain barrier, and supply glucose to most cells. Glut-2 provides glucose to the liver and pancreatic cells. Glut-3 is the main transporter in neurons, whereas Glut-4 is primarily expressed in muscle and adipose tissue and regulated by insulin. Glut-5 transports fructose in intestine and testis. Glut-6/SLC2A6, originally described as glut-9 has been reassigned as Glut-6. Glut-6 (human 507 aa; ~ 48 kDa; chromosome 9q34) is most closely related to Glut-8 (~45% homology). It is highly expressed in brain, spleen, and peripheral leukocytes. Glut-7, expressed in liver and other gluconeogenic tissues, mediates glucose flux across endoplasmic reticulum membrane. Glut-8 is found in adult testis and placenta. Human Glut-9 is expressed in spleen, peripheral leucocytes and brain. Human Glut-10 (541 aa, chromosome 20q13.1; ~30-35% homology with Glut-3 and Glut-8) has been identified as a candidate gene for NIDDM susceptibility. It is widely expressed with highest levels in liver and pancreas. Glut-11 (496 aa, chromosome 22q11.2; ~41% identity with Glut-5) is expressed in heart and skeletal muscle. Glut-12 (human 617 aa, , monkey 621 aa;~ 50 kDa; ~30% homology with Glut-4 and 40% with Glut-10) is expressed in skeletal muscle, adipose tissue, and small intestine . Glut-13 or proton myo-inositol transporter (HMIT; human 629 aa, rat 618 aa, ~75-90 kDa/67 kDa protein) is highly expressed in glial cells and some neurons. Glut-13 transport activity was specific for myo-inositol.
