Presentation: 100ug affinity pure antibody LYOPHILIZED from 1mg/ml PBS stock with 0.1% BSA and 0.02% merthiolate).

Reconstitution: Add 0.1ml sterile DI water or buffer. Let set at least 30 minutes at room temp. Mix well. If desired, centrifuge for 1-2 minutes at 1000rpm to concentrate material in vial. It is not recommended to store diluted solutions. Avoid repeated freeze and thaw.

Reconstitution: All peptide/antibody received in solution or reconstituted from Lyophilized vials should be stored frozen at -20oC or below in suitable aliquots. It is not recommended to store diluted solutions. Avoid repeated freeze and thaw.

Specificity: The antibody has detected GLUT-4 in rat skeletal muscle, heart and adipose tissues. It also recognizes the mouse and human GLUT-4 but shows no cross-reactivity to Hep G2 type transporter from rat brain or other Glucose transporters. The mouse Glut-4 peptide (aa 499-510) sequence shows 100% identity with the corresponding rat and human (aa 498-509) sequences.

USE: Western Blotting (1-10 ug/ml for affinity pure using Chemiluminescence technique). The antibody identified a major band at about 40-43kDa using the GLUT-4 positive control prepared from purified rat adipocyte membranes.\

ELISA (1:10K; using 1ug/ml control peptide/well).

Immunoprecipitations:not tested

Histochemistry & Immunofluorescence:We recommend the testing of affinity purified antibody at 2-10 ug/ml on frozen sections of rat tissues and 3T3 L1 adipocytes.

IMMUNIZING ANTIGEN Partially purified GLUT-4

PURIFICATION METHOD Affinity purified on a Sepharose-4B CNBr linked peptide column

SPECIFICITY The affinity purified antisera is specific for human glucose transporter-4 from muscle and adipose tissue. The antisera does not cross-react with GLUT-1. The molecular weight of the glucose transporter precipitated by the serum is approximately 46 kDa.

PRESENTATION The monoclonal is presented at a concentration of 100µg/ml in phosphate buffered saline containing 10mM sodium azide. We recommend that each laboratory determine an optimum working titre for use in its particular application.

For use within 1 month of purchase store at +4oC, for long term storage aliquot antibody into small volumes and store at -20oC.
