Construct: truncated cDna coding for the extracellular part of human TPO. Transmembrane and intracytolplasmic parts of the molecule are removed and replaced by a hexa-histidine purification tag at the C terminus. After cleavage of the secretory leader presequence, the mature recombinnat TPO has a length of 834 amino acids.

Purity: >95% by SDS-Page analysis, Densitometric analysis. Probed with monoclonal anti-6-His tag.

Immunreactivity: Elisa standard test, checker board analysis of positive/negative sera panels including international referece serum NIBSC 66/387)

MW: apparent=101kDa (calculated 92,872)

Isoelectric point: pH 6.2 (calculated)

Storage: -70 DEG C. At first thaw, tighten cap, centrifuge 1 minute 1000 rpm to concentrate material in vial. Store upright. Avoid frequent warming cycles. Recommend making working aliquots and store at -70 DEG C. Use of reducing sulfhydryl reagents and denaturing detergents will result in destruction of the conformational epitope.

Dilution buffers: recommend ion strength approx 150mM, neutral to slightly alkaline pH and 20% glycerol as cryoprotective agent.

Elisa: approx coating concentration 0.15-0.4ug/ml (titrate for each use)
