Itroduction:
The Pelikine Tool set has been developed to
complement CLB's successful line of Pelikine Compact Elisa kits. The latter have been designed
to combine the ease of a ready-to-use kit with the economy of a do-it-yourself
assay. Thus, the user of our Pelikine Compact kits
only has to make a few simple buffer solutions and Elisa reagents. The Pelikine Tool set contains the chemicals and solutions to
be used in the Pelikine Compact kits: coating and washing
buffers, as well as substrate and stop solutions. Now you are certain of
high-quality solutions, to obtain optimal Elisa results and by using
these ready-made components, you save time and prevent mistakes as well.

II. Storage and Stability:
The Pelikine Tool set should be stored at 2-8'C. The performanceof the kit is guaranteed until the expiration date shown on the bag label.
III. Contents of the Set:
The Pelikine Tool set contains material sufficient for one Pelikine compact kit (288 tests), reagents provided are:

IV. Precautions For Use
1) The Pelikine Tool set is intended for research purposes only.
2) Only use the reagents supplied with the set, do not mix re agents from different kit lots.
3) Do not add any preservative to the supplied reagents, they may have direct or indirect effects on the final colour develop ment of the HRP-substrate system.
4) The washing buffer contains thiomersal (0.001% w/v) and may be toxic upon ingestion, inhalation or skin contact. Avoid con tact of skin, eyes or clothing with dilution, washing or substrate buffer. In case of contact, wash skin or eyes with water and consult a physician.
5) Avoid any skin or eye contact with TMB substrate solution or stop solution provided with this set. In case of contact,wash skin or eyes with water and consult a physician.
6) Do not expose the TMB substrate solution to strong light during incubation or storage. The TMB substrate solution must be colourless when used; if the solution turns blue, it must be replaced.
7) TMB substrate solution or stop solution should not be in contact with metals or metal ions, 10 avoid unwanted colour formation.
V. Additional Materials Required:
- Distilled or deionized water.
- Beakers, flasks,cylinders necessary for preparation (and storage)of prepared buffers.
VI. Preparation of Buffers:
Coating buffer:
Dissolve the contents of one capsule in 100 ml of distilled water. Wait for 5 minutes, mix contents and the working- strength coating buffer is ready for use.
The prepared buffer can be stored in a closed container up to one week at 2-8'C.
Bring buffer to room temperature (18-25'C) and apply buffer as described in your Pelikine compact kit insert.
PBS solution:
Dissolve one tablet in 200 ml of distilled water.Wait for 5 minutes, mix contents and the working-strength phosphate buffered saline (PBS) solution is ready for use.
The prepared buffer can be stored in a closed container up to one week at 2-8'C.
Bring buffer to room temperature (18-25'C) and apply buffer as described in your Pelikine compact kit insert.
Wash buffer:
Prepare a working-strength solution by adding 50 ml of the wash buffer concentration (total content of the bottle) to 950 ml distilled water. The working-strength solution wash buffer can be stored up to 2 months at 2-8'C.
Bring buffer to room temperature (18-25'C) and apply buffer as described in your Pelikine compact kit insert.
VII. Substrate and Stop Solutions:
TMB substrate solution
The supplied substrate solution is a ready-for-use mixture of 3, 3',5,5'-tetramethylbenzidine (TMB) and hydrogen peroxide.Protect substrate solution from prolonged exposure to light.
Bring TMB substrate solution to room temperature (18-25'C) and apply as described in your Pelikine compact kit insert.
Stop solution:
The supplied stop solution is a ready-for-use sulphuric acid solution (0.18M) in water.
Bring stop solution to room temperature (18-25'C) and apply as described in your Pelikine compact kit insert.
VIII. Material Safety Data Sheet:
Hazardous Ingredients
3,3',5,5'-Tetramethylbenzidine may be harmful inhalation, ingestion, or skin absorption. May cause irritation. To our best knowledge the chemical, physical and toxicological properties have not been thoroughly investigated. CAS No. 54827-17-7.
Thiomersal may be toxic upon ingestion, inhalation or
skin contact. Avoid contact of skin, eyes or clothing with dilution, washing or
substrate buffer. In case of contact, wash skin or eyes with water and consult
a physician. CAS No. 54-64-8
Sulphuric Acid: 0.18 M H2SO4 in water. In case of
contact, wash skin or eyes with water and consult a physician. CAS No.
7664-93-9.
