Q: What effect does coagulation of a blood sample have on cytokine assays?
A: This is a very important issue. The outcome of an assay can be largely
dependent upon the method of sample collection, the use of anti-coagulants,
the processing of the sample and the storage conditions. This will restrict
the use of an assay to only certain types of samples. An example is given
in Fig.3, showing that storage of blood, particularly at room temperature
and higher, may induce artificially high levels of IL-8.
Q: When I use PBS prepared from tablets, my cytokine ELISAs give poor results...
A: This a a familiar problem. Some commercially available buffer tablets
contain polyethylene glycol (PEG) as a tableting agent. PEG strongly decreases
the coating efficiency of antibody to the microplate. It is advisable to
check the contents of tableted PBS or to use a powdered preparation.
Q: What are the effects of incubation temperature and shaking on the results
of a cytokine ELISA?
A: Within the usual range, the incubation temperature has little effect.
However, shaking (orbital plate shaker,700 rpm) may give a marked increase
of signal.

Q: In my ELISAs often all wells of one column show a lower absorbancy. How
come?
A: This is a common problem, illustrated in Fig.4. It is caused by a faulty
pipette channel or leaky tips. Remedies: always check your tip attachement
and for every new run change the orientation of the multichannel pipette
sequence, so averaging out any systematic error.
