DATA SHEET:recombinant MURINE LEPTIN
-Leptin (Anti-Obesity Protein) is the protein product of the ob (obese) ene in mice and seems to be involved in appetite control. The ob gene is expressed in adipose tissue and is thought to regulate the body's fat stores. Mice with the ob/ob genotype also develop a form of diabetes similar to type II (non insulin dependent) diabetes. Murine Leptin is a 16 kDa protein containing 147 amino acid residues. -(human leptin also available)

Purity: >95% by SDS-Page analysis and N-terminal sequence analysis. Endotoxin level is less than 0.1ng per ug
(1Eu/ug) of murine leptin.
Reconstitution: The lyophilized leptin is poorly
soluble at** neutral pH and WILL NOT dissolve in a phosphate buffer.
Reconstitute the protein to a concentration of less than 5mg/ml using a 5-10mM
Na citrate pH4.0 buffer. Allow to set at least 30 minutes, tighten cap, rotate
and mix well. If desired, can centrifuge for 1-2 minute at
1000rpm to concentrate material in vial. Once the protein is completely
dissolved, the pH can be readjusted to the physiological range by adding a
titrated amount of 1M stock of Tris pH 8.5.
Storage: The lyophilized powder is stable at room temperature but it is best
stored desiccated below 0 Deg C. Reconstituted Leptin
should be stored in working aliquots at -20 DEG C. AVOID FREQUENT FREEZE THAW
CYCLES
Reactivity: This murine leptin
has been shown to be biologically active in two different mouse obesity models,
ob/ob and NZO. Both strains of mice were treated via intraperitoneal
injection once daily at a dose of 5ug leptin/gm of
body weight for 14 days. Significant effects on body weight, food consumption
and plasma glucose levels were observed to saline treated controls.
