-BAFF, a member of the TNF family of ligands, is expressed in T cells,
macrophages, monocytes and dendritic cells. BAFF is involved in stimulation
of B and T cell function, and is an important survival and maturation factor
for peripheral B cells. BAFF signals through three different TNF receptors
TACI, BCMA and BAFF-R. The human BAFF gene codes for a 285 amino acid type
II transmembrane protein containing a 46 amino acid cytoplasmic domain, a
21 amino acid transmembrane domain, and a 218 amino acid extracellular domain.
This recombinant human soluble BAFF is a 153 amino acid polypeptide (17.0
kDa), which contains the TNF-like portion of the extracellular domain of
BAFF

Purity: >95% by SDS-Page analysis. Endotoxin level is less than 0.1ng
per ug (1EU/ug) of human BAFF.
Reconstitution: The lyophilized BAFF is soluble in water and most aqueous
buffers. The lyophilized powder should first be quick spun in vial, then
reconstituted in water to a concentration of 0.1-1.0mg/ml. Allow to set at
least 30 minutes at 4 DEG C. If desired, can centrifuge for 1 minute at 1000
rpm to concentrate material in vial. This solution can be diluted into water
or other buffered solutions or stored at -20 DEG C for future use.
Note:If this material is not to be used for coating or labeling (or other
applications requiring carrier free material), the addition of 0.1% BSA to
dilution buffers or dilution in sterile culture media is recommended to enhance
stability and minimize absorption to vial. Ideally, maintain stock
>50ug/ml.
Storage: The lyophilized powder, though stable at room temperature, is best
stored dessicated at -20 DEG C. Reconstituted BAFF should be stored in working
aliquots at -20 DEG C. Avoid Frequent Freeze-THAW Cycles.
Reactivity:
Assay#1: The ED50 as determined by the dose dependent stimulation of IL-8 production by Human PBMC is less than 10 ng/ml. Results may vary with PBMC donors.
Assay#2: Determined by a mouse splenocyte survival assay. Ed50 approx 0.5-2ug/ml
