Description: FasL is a type II membrane protein belonging to the TNF family of cytokines. FasL induces apoptosis when it binds to cells expressing the Fas antigen. FasL was first discovered on the cell surface of T cells and NK cells. Expressed on the cell surface of activated T cells, FasL kills T cells and activated B cells leading to down-regulation of the immune responce. The full length of FasL contains 281 amino acid residues, consisting of the transmembrane domain, extracellular domain and the cytoplasmic region. A metalloproteinase cleaves the membrane-bound FasL to produce soluble FasL (sFasL), which exists as a homotrimer. The sFasL shows lower cytotoxic activity than membrane bound FasL. This recombinant Fas Ligand is a soluble 19.9 kDa protein containing 175 amino acid residues, comprising of only the extracellular domain of FasL.

Reconstitution: The lyophilized sFAS-L is soluble in most aqueous buffers. The lyophilized protein should be reconstituted in sterile DI water to a maximum concentration of 0.1-1mg/ml. Allow to set at least 30 minutes at 4 DEG C, tighten cap and rotate to reconstitute any material on sides of vial. If desired, you can centrifuge at 1000 rpm for 1 minute to concentrate volume in bottom of vial. This solution can be diluted into other buffered solutions containing carrier protein or medium and stored at 4 DEG C for up to one week or at -20 DEG C for future use.
Note:If this material is not to be used for coating or labeling (or other applications requiring carrier free material), the addition of 0.1% BSA to dilution buffers or dilution in sterile culture media is recommended to enhance stability and minimize absorption to vial. Ideally, maintain stock >10ug/ml.
Storage: The lyophilized powder is stable at room temperature for a few weeks but it is best stored desiccated at -20 DEG C. Reconstituted sFASl-L should be stored in working aliquots at -20DEG C. AVOID FREQUENT FREEZE THAW CYCLES.
Reactivity: Determined by it's ability to induce cytotoxicity in Jurkat cells in the absence of any cross-linking. The ED50 for this effect is < 10ng/ml, corresponding to a specific activity of > 100K units/mg.
