DATA SHEET:recombinant MURINE TUMOR NECROSIS FACTOR-A

-TNF-alpha is a pleiotrophic pro-inflammatory cytokine secreted by various cells including adipocytes, activated monocytes, macrophages, B cells, T cells and fibroblasts. It belongs to the TNF family of ligands and signals through two receptors, TNFR1 and TNFR2. TNF-alpha is cytotoxic to a wide variety of tumor cells and is an essential factor in mediating the immune response against bacterial infections. TNF-alpha also plays a role in the induction of septic shock, auto immune diseases, rheumatoid arthritis, inflammation, and diabetes. This recombinant murine TNF-alpha is a soluble 156 amino acid protein (17.3 kDa) which corresponds to C-terminal extracellular domain of the full length transmembrane protein.

Reconstitution: The lyophilized TNF-A should be reconstituted in sterile water to a concentration of 0.1-1mg/ml. Allow to set at least 30 minutes at 4 DEG C, rotate/mix well. This solution can be diluted into other buffered solutions or stored at 4 DEG C for up to 1 week or at -20 DEG C for future use. For most in-vitro applications, TNF-A exerts its biological activity in the range of 0.05 to 20.0 ng/ml.

Note:Note:If this material is not to be used for coating or labeling (or other applications requiring carrier free material), the addition of 0.1% BSA to dilution buffers or dilution in sterile culture media is recommended to enhance stability and minimize absorption to vial.

Storage: The lyophilized powder is stable at room temperature for a few weeks but it is best stored desiccated at -20 DEG C. Reconstituted TNF-A should be stored in working aliquots at -20DEG C. AVOID FREQUENT FREEZE THAW CYCLES.

Reactivity: The ED50 as determined by the cytolysis of murine L929 cells in the presence of Actinomycin-D is less than 0.1 ng/ml. This yields a specific activity of greater than 1 X 107 units/mg.
