DATA SHEET:recombinant Human Stem Cell Factor SCF
Stem Cell Factor (SCF) is a hematopoietic growth factor that exerts its activity at the early stages of hematopoiesis. SCF stimulates the proliferation of myeloid, erythroid and lymphoid progenitors in bone marrow cultures and has been shown to act synergistically with colony stimulating factors. Human SCF is an 18.4 kDa protein containing 164 amino acid residues.

Supplied: Lyophilized protein from 10mM acetic acid with no
additives. Endotoxin level is less than 0.1ng
per ug (1 EU/microgram) of human SCF.

Purity: >98% by SDS-Page and HPLC analysis.
Reconstitution: Before opening, tighten cap and tap vial on table or quick spin
to pull lyophilized powder to bottom of vial. The lyophilized powder should be
reconstituted in 10mM acetic acid to a conc of
0.1-1mg/ml. Allow to set at least 30 minutes at 4 DEG
C, rotate, mix well. If desired, centrifuge for 1-2 minutes at 500-1000 rpm to
concentrate material in vial. This solution can be diluted into other buffered
solution containing carrier protein or medium containing serum. These solutions
should be stored at 4 DEG C for up to 1 week or at -20 DEG C for future use.
For most in-vitro applications, SCF exerts its biological activity in the
concentration range of 0.2 to 20.0 ng/ml.
Storage: The lyophilized powder, though stable at room temperature for a few weeks, is best stored dessicated at -20 DEG C. Reconstituted SCF should be stored in working aliquots at -20 DEG C. Avoid Frequent Freeze-THAW Cycles.
Reactivity: The ED50 as determined by the dose dependent stimulation of TF-1
cells was found to be <2ng/ml. This corresponds to a specific
activity of > 5 X 10 E05 (500K)units/mg.

is a single, non-glycosylated polypeptide chain
containing 164 amino acids and having a molecular mass of 18409
Appearance: Sterile Filtered White lyophilized (freeze-dried) powder.

Endotoxin: Less than 0.1 ng/µg (IEU/µg) of rmSCF.
Formulation: Lyophilized from a concentrated (1mg/ml) solution in water containing 10mM acetic acide.
Purity: Greater than 98.0% as determined by:
(a) Analysis by RP-HPLC.
(b) Anion-exchange FPLC.
(c) Analysis by reducing and non-reducing SDS-PAGE Silver Stained gel.
Amino acid sequence: The sequence of the first five N-terminal amino acids was determined and was found to be
Met-Glu-GLY-ILE-CYS.
Dimers and aggregates: Less than 1% as determined by silver-stained SDS-PAGE gel analysis.
Solubility: It is recommended to reconstitute the lyophilized rSCF in sterile 18M-cm H2O not less than 100µg/ml, which can then be further diluted to other aqueous solutions.
Stability: Lyophilized rSCF although stable at room temperature for 3 weeks, should be stored desiccated below -18 C. Upon reconstitution rSCF should be stored at 4 C between 2-7 days and for future use below -18 C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Please prevent freeze-thaw cycles.
Biological Activity: rSCF is fully biologically active when compared to standard. The ED50 as determined by the dose-dependant stimulation of Human TF-1 cells eis less then 2ng/ml, corresponding to a Specific Activity of 100K IU/mg.
Protein content: Protein quantitation was carried out by two independent methods:
1. UV spectroscopy at 280 nm using the absorbency value of 0.52 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics).
2. Analysis by RP-HPLC, using a standard solution of SCF as a Reference Standard.
