-Defensins (alpha and beta) are cationic peptides with a broad spectrum of antimicrobial activity that comprise an important arm of the innate immune system. The alpha-defensins are distinguished from the beta-defensins by the pairing of their three disulfide bonds. To date, four human beta-defensins have been identified; BD-1, BD-2, BD-3 and BD-4. Beta-defensins are expressed on some leukocytes and at epithelial surfaces. In addition to their direct antimicrobial activities, they are chemoattractant towards immature dendritic cells and memory T cells. The beta-defensin proteins are expressed as the C-terminal portion of precursors and are released by proteolytic cleavage of a signal sequence and, in the case of BD-1 (36 a.a.), a propeptide region. Beta-defensins contain a six-cysteine motif that forms three intra-molecular disulfide bonds. Beta-Defensins are 3-5 kDa peptides ranging in size from 33-47 amino acid residues. This recombinant Human BD-2 is a 4.3 kDa protein containing 41 amino acid residues.

Reconstitution: Lyophilized BD-2 should be reconstituted in water to a
concentration of 0.1-1mg/ml. Allow to set at least 30 minutes at 4 DEG C,
mix well. If desired, tighten cap and centrifuge for 1 minute at 1000rpm
to concentrate material in vial. This solution can be diluted into other
buffered solutions or stored at 4 DEG C for up to 1 week or at -20 DEG C
for future use. -Note:If this material is not to be used for coating or labeling
(or other applications requiring carrier free material), the addition of
0.1% BSA or HSA to dilution buffers or dilution in sterile culture media
is recommended to enhance stability and minimize absorption to vial. Ideally,
maintain stock >20ug/ml.
Storage: The lyophilized powder is stable at room temperature for a few weeks
but it is best stored desiccated at -20 DEG C. Reconstituted BD- 2 should
be stored in working aliquots at -20 DEG C. AVOID FREQUENT FREEZE THAW
CYCLES.
Reactivity: Determined by its ability to chemoattract immature human dendritic
clles using a concentration range of 10.0-100.0 ng/ml.
