DATA SHEET:recombinant Human Lymphotactin

-Human Lymphotactin is produced by activated progenitor T cells (pro-T cells) and represents a novel addition to the chemokine family of proteins, characterized by specific chemotactic activity for lymphocytes. This newly discovered chemokine is an 11 kDa polypeptide that shares sequence similarities with members of both the Rantes/sis (C-C) and the IL-8 (C-X-C) families of chemokines. At a concentration of 10-10 M, lymphotactin was found to induce chemotaxis in CD4+CD8+ depleted thymocytes, while a concentration of 10-8 M was required to induce a response in thymic CD8+ depleted cells, as well as in T cell depleted spleen cells, bone marrow cells, and lymph node cells. Neither monocytes, neutrophils, nor even thymic CD4+CD8+ cells respond to the chemotaxis activity of lymphotactin. Recombinant human lymphotactin is a 10 Kda protein containing 92 amino acid residues.

Purity: >98% by SDS-Page analysis. Endotoxin level is less than 0.1ng per ug (1EU/ug)of   lymphotactin.

Reconstitution: The lyophilized lymphotactin should be re to a concentration of 0.1-1.0mg/ml . Allow to set at least 30 minutes at 4 DEG C , tighten cap and rotate to reconstitute any material on sides of vial. If desired, you can centrifuge at 1000 rpm for 1 minute to concentrate volume in bottom of vial. This solution can be diluted into buffer containing carrier protein or medium containing serum and stored at 4 DEG C for up to 1 week or at -20 DEG C long term.

Note:If this material is not to be used for coating or labeling (or other applications   requiring carrier free material), the addition of 0.1% BSA to dilution buffers or   dilution in sterile culture media is recommended to enhance stability and minimize absorption to vial. Ideally, maintain stock >10ug/ml.

Storage: The lyophilized powder is stable at room temperature for a few weeks but it is   best stored desiccated at -20 DEG C. Reconstituted material should be   stored at 4 DEG C (1 week) or in working aliquots at -20 DEG C. AVOID   FREQUENT FREEZE THAW CYCLES.

Reactivity: The biological activity of this rec human lymphotactin was determined by a   chemotaxis assay using human T cells. Significant chemotaxis was achieved using   a concentration of 100ng/ml..
