DATA SHEET:recombinant Human Thrombopoietin

-TPO is a lineage specific growth factor, produced in the liver, kidney and skeletal muscle. It stimulates the proliferation and maturation of megakaryocytes, and promotes increased circulating levels of platelets in vivo. TPO signals through the c-mpl receptor and acts as an important regulator of circulating platelets. Human and murine TPO exhibits cross-species reactivity. This recombinant human TPO is a fully biologically active 174 amino acid polypeptide (18.6 kDa), which contains the erythropoietin-like domain of the full length TPO protein.

Purity: >98% by SDS-Page analysis. Endotoxin level is less than 0.1ng per ug (1EU/ug) of TPO.

Reconstitution: The lyophilized TPO should be reconstituted in 5mM Tris pH 8.0 to a concentration of 0.5-1.0 milligram/ml . Allow to set at 4 DEG C for at least 30 minutes, tighten cap, mix well. If desired, centrifuge for 1-2 minutes at 1000 rpm to concentarte material in vial. This solution should be diluted into buffer containing carrier protein or medium containing serum and stored at 4 DEG C for up to 1 week or at -20 DEG C for long term use.

Note: For material that will not be used for coating or labelling (or other applications requiring carrier free material), we recommend adding 0.1% protease free BSA/HSA or media to any dilution buffers. Maintain stock >10ug/ml.

Storage: The lyophilized powder is stable at room temperature for a few weeks but it is best stored desiccated at -20 Deg C. Reconstituted material should be stored at 4 DEG C or in working aliquots at -20 DEG C. AVOID FREQUENT FREEZE THAW CYCLES.

Reactivity: The ED50 as determined by the dose dependent stimulation of human M07e cells was found to be < 1.0ng/ml corresponding to a specific activity of > 1 million units/mg.
