DATA SHEET:recombinant MURINE STEM CELL FACTOR

-SCF is a hematopoietic growth factor that exerts its activity by signaling through the c-Kit receptor. SCF and c-Kit are essential for the survival, proliferation and differentiation of hematopoietic cells committed to the melanocyte and germ cell lineages. Human SCF manifests low activity on murine cells, while murine and rat SCF are fully active on human cells. This recombinant murine SCF is an 18.3 kDa polypeptide containing 164 amino acid residues, which corresponds to the sequence of the secreted soluble form of SCF.

Supplied: Lyophilized powder from a 10mM Sodium citrate, pH4.0  or 10mM Sodium Phosphate pH6.5 solution . No carrier/additives.

Purity: >95% by SDS-Page analysis and HPLC analysis. Endotoxin level is less than 0.1ng per ug (1EU/ug) of SCF.

Reconstitution: The lyophilized mSCF should be reconstituted in sterile DI water to a maximum concentration of 0.1- 1mg/ml. Allow to set at least 30 minutes at 4 DEG C , tighten cap and rotate to reconstitute any material on sides of vial. If desired, you can centrifuge at 1000 rpm for 1 minute to concentrate volume in bottom of vial. This solution can be diluted into other buffered solutions containing a carrier protein (as 0.1% BSA) or media containing serum or stored at 4 DEG C for up to 1 week or at -20 DEG C for future use. For most in-vitro applications, SCF exerts its biological activity in the range of 1.0 to 100ng/ml.

Note:If this material is not to be used for coating or labeling (or other applications requiring carrier free material), the addition of 0.1% BSA or dilution in sterile culture media is recommended to enhance stability and minimize absorption to vial.


Storage: The lyophilized powder is stable at room temperature for a few weeks but it is best stored desiccated at -20 DEG C. Reconstituted SCF should be stored in working aliquots at -20 DEG C. AVOID FREQUENT FREEZE THAW CYCLES.

The ED50 was determined by the dose dependent stimulation of the proliferation of human TF-1 cells is <10ng/ml, corresponding to a specific activity of >100K units/mg.

Old: Reactivity: The ED50 as determined by the dose-dependent proliferation of the human M07e cell line is <20ng/ml, corresponding to a specific activity of >5 X 10 E04 units/mg.

Description: Recombinant murine SCF produced in E.Coli is a single, non-glycosylated polypeptide chain containing 164 amino acids and having a molecular mass of 18309 Dalton. The SCF is purified by proprietary chromatographic techniques.

Appearance: Sterile Filtered White lyophilized (freeze-dried) powder.

Endotoxin: Less than 0.1 ng/µg (IEU/µg) of rmSCF.

Formulation: Lyophilized from a concentrated (1mg/ml) solution in water containing 5mM Sodium phosphate buffer pH=6.8.

Purity: Greater than 95.0% as determined by:

(a) Analysis by RP-HPLC.

(b) Anion-exchange FPLC.

(c) Analysis by reducing and non-reducing SDS-PAGE Silver Stained gel.

Amino acid sequence: The sequence of the first five N-terminal amino acids was determined and was found to be Met-Lys-Glu-Ile-Cys.

Dimers and aggregates: Less than 1% as determined by silver-stained SDS-PAGE gel analysis.

Solubility: It is recommended to reconstitute the lyophilized rmSCF in sterile 18M-cm H2O not less than 100µg/ml, which can then be further diluted to other aqueous solutions.

Stability: Lyophilized rmSCF although stable at room temperature for 3 weeks, should be stored desiccated below -18 C. Upon reconstitution rmSCF should be stored at 4 C between 2-7 days and for future use below -18 C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Please prevent freeze-thaw cycles.

Biological Activity: rmSCF is fully biologically active when compared to standard. The ED50 as determined by the dose-dependant stimulation of Human MO7e cell line is less then 20 ng/ml, corresponding to a Specific Activity of 200K IU/mg.

Protein content: Protein quantitation was carried out by two independent methods:

1. UV spectroscopy at 280 nm using the absorbency value of 0.52 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics).

2. Analysis by RP-HPLC, using a standard solution of SCF as a Reference Standard.
