DATA SHEET:recombinant Human INTERLEUKIN-13

IL-13 is an immunoregulatory cytokine produced primarily by activated Th2 cells, and also by mast cells and NK cells. Targeted deletion of IL-13 in mice resulted in impaired Th2 cell development and indicated an important role for IL-13 in the expulsion of gastrointestinal parasites. IL-13 exerts anti-inflammatory effects on monocytes and macrophages and it inhibits the expression of inflammatory cytokines such as IL-1beta, TNF-alpha, IL-6 and IL-8. IL-13 has also been shown to enhance B cell proliferation and to induce isotype switching resulting in increased production of IgE. Blocking of IL-13 activity inhibits the pathophysiology of asthma. Human and murine IL-13 is cross-species reactive. This recombinant human IL-13 is a 12.5 kDa protein consisting of 114 amino acid residues.

Reconstitution: Lyophilized IL-13 should be reconstituted in 20mM HCL to a concnetration of 0.1-1mg/ml(Note: The material is slow to dissolve). Allow to set at least 30 minutes at 4 DEG C, mix well. If desired, centrifuge 1 minute 1000rpm to concentrate material in vial. This solution can be diluted into other buffered solutions or stored at 4 DEG C for up to 1 week or at -20 DEG C for future use. For most in-vitro applications, IL-13 exerts its biological activity in the range of 0.1 to 10.0ng/ml.

Note:If this material is not to be used for coating or labeling (or other applications requiring carrier free material), the addition of 0.1% BSA to dilution buffers or dilution in sterile culture media is recommended to enhance stability and minimize absorption to vial. Ideally, maintain stock >50ug/ml.

Storage: The lyophilized powder is stable at room temperature for a few weeks but it is best stored desiccated at -20 Deg C. Reconstituted IL-13 should be stored in working aliquots at -20DEG C. AVOID FREQUENT FREEZE THAW CYCLES.

Reactivity: This product is fully biologically active as measured by the dose dependent proliferation of TF-1 cells. The ED50 was less than 1.0 ng/ml, corresponding to a specific activity of >1 million units per mg.
