Epidermal Growth Factor (EGF) is a potent growth factor that stimulates the
proliferation of various epidermal and epithelial cells. Additionally, EGF
has been shown to inhibit gastric secretion, and to be involved in wound
healing. EGF signals through a receptor known as c-erbB, which is a class
I tyrosine kinase receptor. This receptor also binds with TGF-alpha and VGF
(vaccinia virus growth factor). This recombinant human EGF is a 6.2 kDa globular
protein containing 53 amino acid residues, including 3 intramolecular
disulfide-bonds.

Reconstitution:The lyophilized EGF (which is soluble in water and most aqueous
buffers) should be reconstituted in steriel water to a concentration of 1mg/ml.
Allow to set at least 30 minutes (suggest tightening cap and rotating. Centrifuge
1000 rpm for 1 minute to concentrate material in vial to pull any liquid
of sides of vial). This solution can be diluted into other buffered solutions
or stored at -20 DEG C for future use. For most in- vitro applications, EGF
exerts its biological activity in the range of 0.5 to 25.0 ng/ml.
Storage: The lyophilized powder is stable at room temperature but it is best
stored desiccated below 0 Deg C. Reconstituted EGF should be stored in working
aliquots at -20DEG C. AVOID FREQUENT FREEZE THAW CYCLES.
Reactivity: This EGF is fully biologically active when compared to standards.
The ED50 determined by the dose-dependent stimulation of thymidine uptake
by Balb-c 3T3 cells is <0.1ng/ml, corresponding to a specific activity
of > 1 X 10 X E07 units/mg.

Description: Recombinant Human EGF produced in E.Coli is a single, non-glycosylated polypeptide chain containing 53 amino acids and having a molecular mass of 6222 Dalton. The rHuEGF is purified by proprietary chromatographic techniques.

Appearance: Sterile Filtered White lyophilized (freeze-dried) powder.
Formulation: Lyophilized from a concentrated (1mg/ml) solution in water containing no additives.
Endotoxin: Less than 0.1 ng/µg (IEU/µg) of rHuEGF
Purity: Greater than 98.0% as determined by:
(a) Analysis by RP-HPLC.
(b) Anion-exchange FPLC.
(c) Analysis by reducing and non-reducing SDS-PAGE Silver Stained gel.
Amino acid sequence: The sequence of the first five N-terminal amino acids was determined and was found to be Asn-Ser-Asp-Ser-Glu. N terminal methionine has been completely removed enzymatically.
Dimers and aggregates: Less than 1% as determined by silver-stained SDS-PAGE gel analysis.
Solubility: It is recommended to reconstitute the lyophilized rHuEGF in sterile 18M-cm H2O not less than 100µg/ml, which can then be further diluted to other aqueous solutions.
Stability: Lyophilized rHuEGF although stable at room temperature for 3 weeks, should be stored desiccated below -18 C. Upon reconstitution rHuNT-3 should be stored at 4 C between 2-7 days and for future use below -18 C. For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA). Please prevent freeze-thaw cycles.
Biological Activity: rHuEGF is fully biologically active when compared to standard. The ED50 as determined by the dose-dependent proliferation of murine BALB/c 3T3 cells (as measured by 3H thymidine uptake) is <0.1ng/ml corresponding to a specific activity of 1 X 10 E07 Units/mg
Protein content: Protein quantitation was carried out by two independent methods:
1. UV spectroscopy at 280 nm using the absorbency value of 2.858 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics).
2. Analysis by RP-HPLC, using a standard solution of EGF as a Reference Standard.
