-The three mammalian isoforms of TGF-beta, TGF-beta1, TGF-beta2, TGF-beta3, signal through the same receptor and elicit similar biological responses. They are multifunctional cytokines that regulate cell proliferation, growth, differentiation and motility as well as synthesis and deposition of the extracellular matrix. They are involved in various physiological processes including embryogenesis, tissue remodeling and would healing. They are secreted predominantly as latent complexes which are stored at the cell surface and in the extracellular matrix. The release of biologically active TGF-beta isoform from a latent complex involves proteolytic processing of the complex and /or induction of conformational changes by proteins such as thrombospondin-1. TGF-beta2 has been shown to exert suppressive effects on IL-2 dependent T-cell growth, and may also have an autocrine function in enhancing tumor growth by suppressing immuno-surveillance of tumor development. This Recombinant human TGF-beta2 is a 25.0 kDa protein composed of two identical 112 amino acid polypeptide chains linked by a single disulfide bond.* Manufactured using (BTI-Tn-5B1-4) cells under license from the Boyce Thompson Institute for Plant Research, Inc.

Purity: >98% by SDS-Page analysis and HPLC analysis. Endotoxin less than 0.1ng per ug of TGF-B2.

Reconstitution: Human TGF-B2 is very soluble in buffers at pH's below 4. However, at neutral pH's TGF-B2 tends to stick to surfaces of tubes, pipets, etc. This TGF-B2, when reconstituted in water to a concentration of 50ug/ml (ie, 5ug/100ul) will yield a solution of 1mM citrate buffer pH 3.5 that can be stored at room temp or at 4 DEG C for one mnths. Dilution of this stock solution should take into account the stickiness of the protein. It is recommended that further dilutions be made into PBS containing 2mg/ml albumin and stored in working aliquots at -20 DEG C.

Storage: The lyophilized powder is stable at room temperature for a few weeks but it is best stored desiccated at -20 DEG C. Reconstituted stock TGF-B2 (50ug/ml in citrate buffer) can be stored at room temp or 4 DEG C for at least 1 month. More dilute solutions of TGF-B2 in PBS/2mg/ml albumin should be stored at -20 DEG C.AVOID FREQUENT FREEZE THAW CYCLES.

Reactivity: The ED50 was determined by TGF-B2 ability to inhibit the mouse IL-4 dependent proliferation of mouse HT-2 cells is less than 0.2ng/ml, corresponding to a specific activity of >5 X 10 E06 units per milligram.
