recombinant Human Vascular Endothelial Cell Growth Factor (165)
Vascular Endothelial Cell Growth Factor (VEGF) is a secreted homodimeric
protein secreted by a variety of vascularized tissues. The reported activities
of VEGF include stimulation of endothelial cell growth, angiogenesis, and
capillary permeability. Human VEGF is a 38.2 kDa homodimeric protein consisting
of two 165 amino acid polypeptide chains.

Reconstitution: The lyophilized VEGF is soluble in water and most aqueous
buffers. The lyophilized powder should be reconstituted in water to a max concentration
of 0.1-1mg/ml. Allow to set at least 30 minutes at 4 DEG C, tighten cap and
rotate to reconstitute any material on sides of vial. If desired, you can
centrifuge at 1000 rpm for 1 minute to concentrate volume in bottom of vial.
This solution can be diluted into water or other buffered solutions or stored
at 4 DEG C for up to 1 week or at - 20 DEG C for future use. For most in-vitro
applications, human VEGF exerts its biological activity in the concentration
range of 1.0 to 50.0 ng/ml.
Note:If this material is not to be used for coating or labeling(or other applications requiring carrier free material), the addition of 0.1% BSA to dilution buffers or dilution in sterile culture media is recommended to enhance stability and minimize absorption to vial.
Storage: The lyophilized powder is stable at room temperature for a few
weeks but it is best stored desiccated at -20 Deg C. Reconstituted human VEGF
should be stored in working aliquots at -20 DEG C. AVOID FREQUENT FREEZE THAW
CYCLES.

Description: Human Vascular Endothelial Growth Factor VEGF165, a 23 kDa protein consisting of 165 amino acid residues, is produced as a homodimer. VEGF is a polypeptide growth factor and a member of the platelet-derived growth factor family. It is a specific mitogen for vascular endothelial cells and a strong angiogenic factor in vivo. Two high-affinity tyrosine kinase receptors for VEGF165 have been identified, VEGFR-1 (FLT -1), and VEGFR-2 (KDR). Consistent with the endothelial cell-specific action of VEGF165, expression of both receptor genes has been found predominantly but not exclusively on endothelial cells. Expression of VEGFR-1 was also found on human monocytes, neutrophils (PMNs), bovine brain pericytes and villous and extravillous tropho-blasts. In addition to its action as a mitogen it is a potent vasular permeability factor (VPF) in vivo. VEGF165 is also a chemoattractant molecule for monocytes and endothelial cells. 5 different proteins are generated by diffential splicing: VEGF121, VEGF145, VEGF165, VEGF189 and VEGF206. The most abdundant form is VEGF165. Whereas VEGF121 and VEGF165 are secreted proteins, VEGF145, VEGF189 and VEGF206 are strongly cell-associated. The isoforms VEGF145, VEGF165 and VEGF189 bind to heparin with high affinity. VEGF165 is appa-rently a homo -dimer, but preparations of VEGF165 show some heterogeneity on SDS gels, depending on the secretion of different glycosylation patterns. All dimeric forms have similar biological activities but their bio-availability is very different. There is good evidence that heterodimeric molecules between the different isoforms also exists and that different cells and tissues express different VEGF isoforms. The other members of this in-creasing growth factor family are VEGF-B, -C, -D and -E. Another member is the Placenta growth factor PlGF.

VEGF is a potent growth and angiogenic cytokine. It stimulates proliferation and survival of endothelial cells, and promotes angiogenesis and vascular permeability. Expressed in vascularized tissues, VEGF plays a prominent role in normal and pathological angiogenesis. VEGF signals through three receptors; fms-like tyrosine kinase (flt-1), KDR gene product (the murine homolog of KDR is the flk-1 gene product) and the flt4 gene product. Due to its increased acidity, VEGF121 circulates more freely than other VEGF forms, which bind more tightly with vascular heparin sulfates. This recombinant human VEGF121 is a 28.4 kDa disulfide-linked homodimeric protein consisting of two 121 amino acid polypeptide chains.
