Recombinant Human CD137-huIgG (4-1BB) fusion Protein
Background: Human CD137 (4-1BB) is expressed on activated T cells within 24-48 hours of activation. CD137 is a type I membrane protein and a member of the tumor necrosis factor receptor superfamily. CD137 appears to be important for T cell proliferation and survival and induces monocyte activation.

Mw: a soluble 55kd fusion protein consisting of the extracellular (186aa)
domain of human CD137 fused to human IgG1 fc.
Activity: This human CD137 Ig fusion protein blocks
binding of an anti-human CD137 o activated CEM human tunor
cells and also binds to CD137 Ligand on Raji cells.
-can be used in histochemistry (frozen sections)
and flow cytometry
Storage: 2-5 DEG C (DO NOT FREEZE) aliquot and store in working aliquots.

Assay: 500K human Raji cells were washed and
incubated 45 minutes on ice with 80ul of CD137-huIg at 5ug/ml. Cells were
washed twice and incubated with 50ul of secondary antibody (Goat anti-human IgG:FITC) after which they were washed three times, fixed
and analyzed by Facs. Cells stained positive with a
mean shift of 0.48 log10 fluorescent units when compared to a buffer control at
a similar concentration. Binding was blocked when reagent was preincubated 30 minutes with CD137L-muCD8 at a
concentration of 50ug/ml.

Background: Human CD137L (4-1BB Ligand) is a type
II transmembrane protein constitutively expressed by monocytes, B cells and neuroblastoma
cells . Binding of CD137 (4-1BB) to CD137L induces monocyte
activation.

Assay:Cultured human CEM cells were stimulated 1 day in the presence of PMA(10ug/ml) and ionomycin (1uM). 500K cells were washed and incubated for 45 minutes on ice with 80ul of CD137L- muCD8 at 10ug/ml. Cells were washed twice and incubated with anti-mouse CD8A/RPE after which they were washed three times, fixed and analyzed by Facs. Cells stained positive with a mean shift of 1.02 log10 fluorescent units when compared to a buffer control at a similar concentration. Binding was blocked when reagent was preincubated 30 minutes with CD137-huIg at a concentration of 50ug/ml.
