Cytokines, Growth Factors & Chemokines:ANTIBODIES

RDI Divison of Fitzgerald Industries Intl offers a wide line of recombinant growth factors, cytokines and chemokines and antibodies. Since no one antibody works best for all applications (neutralization, blotting, ELISA, etc), we offer many different types of antibodies to help solve this problem. Please inquire for other applications or types of antibodies not listed below.
Rabbit anti-human BDNF:Biotin conjugated (antigen aff pure, conjugated to Biotin, lyophilized, carrier free)
Goat anti-human BDNF (antigen aff pure, lyophilized, no carrier/no preservative)
Goat anti-human BDNF:Biotin conjugated (antigen aff pure, conjugated to Biotin, lyophilized, carrier free)
Produced from sera of rabbits pre-immunized with highly pure (>98%) recombinant hBDNF. Anti-hBDNF specific antibody was purified by affinity chromatography employing immobilized hBDNF matrix.

Formulation : A sterile filtered antibody solution (1mg/ml) was lyophilized from 0.5X PBS pH 7.4.
Stability : The lyophilized antibody is stable at room temperature for at least one month and for greater than a year when kept at -20C.
Reconstitution : When reconstituted in sterile water to a concentration of >0.2 mg/ml the antibody is stable for at least six weeks at 2-4C.
Neutralization Data not available.
Elisa To detect hBDNF by direct ELISA (using 100µl/well antibody solution) a concentration of at least 0.5µg/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with compatible secondary reagents, allows the detection of 0.2-0.4 ng/well of recombinant hBDNF.
Western Blot To detect hBDNF by Western Blot analysis this antibody can be used at a concentration of 0.1-0.2 µg/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant hBDNF is 1.5-3.0 ng/lane, under either reducing or non-reducing conditions.

Optimal dilutions should be determined by user for each application
Storage: Store frozen. Aliquot as undiluted antisera and immediately place at -20 DEG C. At first thaw, tighten cap, centrifuge for 1-2 minutes at 1000 rpm to concentrate material in vial. Stable at least 6 months at -20 DEG C. Avoid repeated freeze/thaw cycles.

Western blotting: Based on western blot results, this antibody shows no cross-reactivity with recombinant human ß -NGF, r recombinant rat ß -NGF, recombinant human NT-3and recombinant human NT-4.
Antiserum was used on fixed-frozen tissue and paraffin embedded tissue. This antibody can be used with appropriate secondary reagents to detect rat and human BDNF. Staining may be done on tissues dissected from animals that were fixed by vascular perfusion with 4% paraformaldehyde/PBS (pH 7.4) and followed by perfusion with a 10% sucrose solution in 0.1 M phosphate BDNF can be detected in naïve animals.
BDNF was found to be a fixation sensitive antigen: post-fixation of tissues with 4% paraformaldehyde may greatly reduce or even abolish BDNF labeling.
