Reconstitution: 100ug: When reconstituted with 70ul distilled water (final volume 100ul),
250ug: when reconstituted with 175ul distilled water (final volume 250ul)
concentration of cytokeratin 14 is 1mg/ml in 30mM Tris/HCL pH 8,
9.5M urea, 2mM DTT, 2mM EDTA, 10mM methylammonium chloride
Storage: At 2-8 DEG C (lyophilized) Store -20 DEG C after reconstitituion Avoid frequent freeze thaw cycles.
-reconstitution to filaments is performed by mixing equimolar amounts of cytokeratins of Type 1 and type II at concentrations of approx 0.5mg/ml, both dissolved in 9.5M urea buffer (as per above formulation) Protofilaments and filament complexes are obtainined by dialyzing the resulting polypeptide stepwise to a concentration of 4M urea and then to low salt concentration (50mM NaCl, 2mM dithiothreitol, 10mM Tris-HCL, pH 7.4).
