Reconstitution:               100ug: When reconstituted with 70ul distilled water (final volume 100ul),

concentration of DESMIN is 1mg/ml in 30mM Tris/HCL pH 8, 9.5M urea, 2mM DTT, 2mM EDTA, 10mM methylammonium chloride

-reconstitution to filaments is performed by mixing equimolar amounts of cytokeratins of Type 1 and type II at concentrations of approx 0.5mg/ml, both dissolved in 9.5M urea buffer (as per above formulation) Protofilaments and filament complexes are obtainined by dialyzing the resulting polypeptide stepwise to a concentration of 4M urea and then to low salt concentration (50mM NaCl, 2mM dithiothreitol, 10mM Tris-HCL, pH 7.4).

Our product cat. no. RDI-PRO62016 (recombinant desmin) also contains low amounts (< 5%) of the 45 kD polypeptide, which is a proteolytic degradation product of desmin and represents the alpha-helical rod piece of the polypeptide. This 45 kD fragment cannot be eliminated totally by our purification methods. When reconstituting the lyophilized product the rod piece is readily soluble whereas the intact polypeptide takes more time to get resolubilized. Whenever the applicant is not investing sufficient time for resolubilization the more soluble 45 kD fragment becomes over-represented in the sample taken for e.g. SDS-PAGE and Western blot analysis. In total homogenates of tissue the 45 kD fragment is found after unsufficient measures against proteolytic degradation.
